Skin scald is a common form of skin wounds, skin wound healing is a matter of inflammation, new tissue formation and tissue reconstruction the complex pathophysiological process. In this process, there are a variety of tissue, cells and extracellular matrix and participate in a variety of cell regulatory factors.Epigallocatechin -3- gallate (epigallocatechin-3-gallate, EGCG) is the main component of green tea polyphenols, antioxidant, anti-tumor, anti-inflammatory response and other characteristics. Studies have shown that EGCG inhibited TNF-α,IL-1βand MMP-1 gene expression and production, and the expression of TIMP-1 gene, and by TNF-α,IL-1β,MMP-1 and TIMP-1 Gene-induced inflammatory response. Previous study in our laboratory showed that EGCG inhibited the healing of skin wounds, decreased tissue levels of IL-1β, suggesting that EGCG may inhibit excessive inflammation occurs the wound and promote wound healing.To study the effect of EGCG on the healing of skin scald and its mechanism, the use of stereology, histology, reverse transcription polymerase chain reaction (reverse transcription polymerase chain reaction, RT-PCR) and ELISA (enzyme-linked immunosorbent assay, ELISA) method of EGCG on skin Scald change and healing during the TNF-α, IL-1β, MMP-1 and TIMP-1 gene expression levels to clinical application of EGCG to improve the skin Scald repair and provide experimental evidence.By stereology and histology can be seen, 0.2 mg / g EGCG treated mice coated first Skin Scald healing, indicating that 0.2mg / g EGCG can promote wound healing in mice. RT-PCR and ELISA also showed that 0.2mg / g EGCG inhibited TNF-α, IL-1βand MMP-1 gene expression, promoting the expression of TIMP-1 gene; and reduce the injury site TNF-αIL-1βand MMP-1 levels. indicating that 0.2mg / g EGCG can area of mice TNF-αIL-1βand MMP-1 levels returned to normal levels quickly, too, that 0.2mg / g EGCG can facilitate scalded skin wound healing in mice.
