The Pharmacodynamics And Mechanism Study Of Yuchang Ning Capsule Treating Ulcerative Colitis

Objective This article took the Yuchangning capsule as the object of study,through establishing the ulcerative colitis model of rats, mainly to research the treatment and mechanism of ulcerative colitis model and its anti-inflammatory effects,so as to provide a reliable Pharmacology basis for the clinical application of Yuchangning capsule.Methods Both TNBS and DSS were used to induce rats ulcerative colitis,through observing the bloody stool,the diarrhea and the food consuming quantity of rats, calculating each rat’s intestinal and spleen weight factor,determinating the MPO content of colons and doing the microscope examionations,and using auricle tumefaction and capillary permeability to study the anti inflammation effection of Yuchangning capsule, immuohistochemistry was used to detect IkB-a protein in colon tissue cells of animals, while TNF-a, IL-1β, IL-6, IL-8cytokines,MDA and SOD content of colonic tissue were measured.Results:These pharmacodynamics’s experiments proved that Yuchangning capsule could treat ulcerative colitis significantly.It could improve rats bloody stool,diarrhea and the food consuming quantity significantly(P<0.05);reduce the intestinal weight coefficient significantly (P<0.05);increase the spleen weight coefficient significantly (P<0.05);reduce the MPO content of intestinal significantly (P<0.01),alleviate the colon histologic damage index significantly(P<0.01),reduce colonic tissue inflammation,lymphocyte infiltration significantly(P<0.01).Yu changning can significantly reduce the auricle tumefaction caused by Dimethylbenzene,inflammation swelling suppression rate achieves mostly44.74%(P<0.01),it can also reduce the capillary permeability,high dose and middle dose may obviously reduce the Evans blue dye quantity which seeps out from the mouse abdominal cavity in the high dose team the Evans blue dye quantity which seeps out from the abdominal cavity (the OD value) the reduction rate achieves39.73%(P<0.01),demonstrated it has the remarkable inhibitory effect to the acetic acid result’s mouse abdominal cavity blood capillary permeability markup;Mechanism experiment proved that Yu changning could significantly ruduce the TNF-a content(P<0.01),IL-1β content(P<0.01),IL-6content(P<0.01)and IL-8content(P<0.01);which indicated that Yuchangning’s anti-inflam matory effection had something to do with its inhabiting inflammatory-inducing factors;increase the SOD content significantly (P<0.01). and reduced the MDA content significantly(P<0.01),which indicated that Yuchangning’s anti-inflammatory effection had something to do with its adjusting radicals disorder and its capacity of clear radicals. It also could significantly inhibit IKB-a protein expressions of animals colonic mucosa (P<0.01),which indicated that Yuchangning’s anti-inflammatory effection had something to do with its reducing the IKB-a protein expression in rats’colonic mucosa.Conclusion:Yuchangning capsule could significantly treat ulcerative colitis which induced by TNBS and the DSS,strengthen bodies’immunity,and has good anti-inflammatory effection. Mechanism experiment proved that Yu changning could interfere IkB-a protein expression,then inhibit the release of inflammatory-inducing factors and adjust radicals disorder to play anti-inflammatory effect.
