ObjectiveThrough detecting the expression of PTCH1gene about Hedgehog signal in patientswith esophageal carcinoma tissues, precancerous tissues and normal tissues, further tounderstand and discuss the expression of PTCH1gene in patients with esophagealcarcinoma occurrence,detecting methods, relationship between PTCH1gene and histology,and relationship between PTCH1gene and pathology.MethodUsing the real-time PCR to detect the expression of PTCH1gene in40cases ofesophageal cancer, the relative tissues and10cases normal esophageal tissues,and dostatistical analysis about tumor stage, clinical pathology material, tumor size in theesophagus, differentiation degree, lymphatic moving or not, age, gender.ResultWe can find the expression of PTCH1gene in every patient with esophagealcarcinoma, precancerous tissues and normal tissues.The relative expression of PTCH1gene in esophageal carcinoma is4.75(2.56-9.09), which is higher than the expression ofPTCH1gene in precancerous tissues0.80(0.4-1.52).The differences between theexpressions have statistical significance(P<0.05).The expression of PTCH1gene inesophageal carcinoma is higher than the expression in normal tissues1(0.592-1.69).Thedifferences between the expressions have statistical significance(P<0.05).The expression inprecancerous tissues and normal tissues do not have difference.The expression of PTCH1gene in patients with esophageal carcinoma is not concerned with esophageal tumor size,lymphatic moving and patients’ age, gender, and it’s concerned with differentiation degreeand pathological category. The expression of PTCH1gene in patients with moderate andhigh differentiated esophageal carcinoma is3.86(3.24±4.48). The expression of PTCH1gene in patients with poorly differentiated is6.09(4.64±7.55).The differences between theexpressions have statistical significance(P<0.05).The expression of PTCH1gene in Esophageal Squamous Cell Carcinoma is5.19(4.32-6.06),which is higher than theexpression of esophageal adenocarcinoma3.00(2.50-3.50).The differences between theexpressions of Esophageal Squamous Cell Carcinoma and esophageal adenocarcinomahave statistical significance(P<0.05).ConclusionIt’s sensitive and specific to detect the expression of PTCH1gene in esophagealcarcinoma using real-time quantitative fluorescence PCR.The expression of PTCH1genein esophageal carcinoma is higher than the expression of PTCH1gene in precanceroustissues and normal tissues.The expression of PTCH1gene in patients with esophagealcarcinoma is not concerned with esophageal tumor size, lymphatic moving and patients’age, gender, and it’s concerned with differentiation degree and pathological category. Theexpression in esophageal squamous cell carcinoma is higher than the expression inadenocarcinoma.The expression in low differentiated esophageal carcinoma is higher thanthe expression in high and moderate differentiated esophageal carcinoma.It suggests thatPTCH1gene is concerned with the development of esophageal cancer and malignantdegree.
