Part â… Objective:To investigate whether chronic partial sleep deprivation affect the sleep pattern in adolescent rats.Methods:Adolescent rats were subjected to sleep deprivation for4h from8:00to12:00each day for consecutive7days. Animals were kept awake by introducing a new object into the cage by tapping lightly the cage or touching the animal with a paintbrush gently. Control animals were undisturbed in cages. EEG and electromyogram were recorded during the whole process of chronic SD procedure.Results:1. Total time of wakefulness reduced and NREM increased after chronic sleep deprivation in light phase.2. EEG power density during NREM and REM sleep for7h of light phase increased after chronic sleep deprivation.3. SWA in light phase increased after sleep deprivation for7days.Conclusion:After chronic partial sleep deprivation for7days and4h per day can influence the structure of sleep and sleep homeostasis.Part â…¡Objectives:To investigate effects of different periods of chronic sleep deprivation on spatial learning and memory and the hippocampal expression of adenosine receptor and PDE after chronic sleep deprivation in rats. To explore the relation between cognition disorder and hippocampal molecular changes caused by chronic sleep deprivation between adolescent and adult rats.Methods:Adolescent and adult male Sprague-Dawley rats were randomly divided into control group(C)sleep deprivation group(SD). SD group were divided into four groups:2h group (SD2),4h group (SD4),6h group (SD6) and8h group (SD8).All the rats were trained for spatial learning in Morris water maze for4days from5:00-7:00in the morning. Then animals were kept awake by introducing a new object into the cage, by tapping lightly the cage or touching the animal with a paintbrush gently. Control animals, which were undisturbed in cages. Morris water maze spatial probe test was performed to test spatial learning and memory of rats after sleep deprivation. Real time RT-PCR method was used to test hippocampal adenosine and PDE4mRNA expression.Results:1. There were no significant differences among all the groups of sleep deprived rats in latency during spatial learning both in immature and adult rats, but the swimming path length significantly increased in the adolescent rats.2. The capacity of learning decreased in adolescent rats while the capacity of memory decreased in adult rats.3. The hippocampal RT-PCR indicated that A1receptor, A2A receptor, and PDE4mRNA increased after chronic sleep deprivation in adolescent rats.Conclusion:After chronic partial sleep deprivation for7days and2h,4h,8h per day could decreased the capacity of learning in adolescent rats.2. The hippocampal real time RT-PCR indicated that A1receptor, A2A receptor, and PDE4mRNA increased after chronic sleep deprivation in adolescent rats.
