| [Objective] To investigate the influence of Testosterone Propionate(TP) on PC12apoptosis induced by Aβ25-35and to explore its mechanism.[Method]PC12cells were evaluated for Aβ25-35alone, TP alone and Aβ25-35plus TP. Apoptosis and its rate were examined by the laser scanning confocal microscope and the flow cytometry. In experiment2, PC12cells were treated TP with different concentrations. ERK1/2phosphorylation level was measured by Western Blot (WB). In the third one, the cells were treated with TP alone or combined with the MEK inhibitor U0126. Seladin-1expression was assessed by WB.[Results] PC12apoptosis induced by AP25-35seriously reduced when pretreated with TP. Further, TP could significantly increase the level of ERK phosphorylation. However, the MEK inhibitor U0126could markedly block the upregulation of seladin-1expression conferred by TP.[Conclusion] Our results demonstrated that testosterone propionate prevented PC12cells from the apoptosis induced by AP25-35. Besides, we also found that TP could promote the activation of MAPK/ERK pathway and increases seladin-1expression. |
