The Expression And Clinical Significance Of Tim-3-gal-9Pathway In Preeclampsia Patients At The Maternal-fetal Interface | | Posted on:2013-09-16 | Degree:Master | Type:Thesis | | Country:China | Candidate:H Y Hao | Full Text:PDF | | GTID:2234330392956591 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | Partâ… The expression of Tim-3-Gal-9pathwayin decidua tissuesã€Objective】Detected the expression of Tim-3and Gal-9in decidua tissues of preeclampsiapatients and normal pregnant women, in order to explore the role of Tim-3-Gal-9pathway in the occurrence of preeclampsia.ã€Methods】1. The expression of Tim-3and Gal-9mRNA in decidua tissues were detected byReal-Time PCR in30preeclampsia patients,30normal pregnant women.2. Immunohistochemical detected the expression of Tim-3and Gal-9in deciduatissues of preeclampsia patients and normal pregnant women, highly precisecolor-image measure system was applied to semi-quantitative analysis.5visualfields of every slice were chosen in the high power lens, the mean optic densitiesof the positive cells were calculated, and then analyze them by statistics.3. Western blot was applied to the detection of Tim-3and Gal-9protein in deciduatissues, then process statistical analysis. ã€Results】1. The relative expression of Tim-3and Gal-9mRNA in preeclampsia patients werehigher than that in normal pregnant women, but statistically insignificant(P>0.05).2. Tim-3and Gal-9were expressed in the decidua tissues both of preeclampsiapatients and normal pregnant women, Tim-3and Gal-9positive cells cytoplasmwere stained brown. Compared to normal pregnant women, the level of Tim-3andGal-9in preeclampsia were significantly higher(P <0.05).3. The relative expression of Tim-3and Gal-9protein in decidua tissues ofpreeclampsia patients were significantl higher than that in normal pregnantwomen (P>0.05).ã€Conclusion】Tim-3and Gal-9were expressed in the decidua tissues both of the preeclampsiapatients and normal pregnant women. The over-expression of Tim-3and Gal-9maybe related to the occurrence of preeclampsia. Partâ…¡ The expression of IFN-γã€IL-10ã€IL-17in peripheralblood plasma of preeclampsiaã€Objective】Comparison of the expression level of interferon-γ (IFN-γ)ã€interleukin-10(IL-10)and interleukin-17(IL-17) in peripheral blood plasma of preeclampsia and normalpregnant women, and then analyzed the correlation of Tim-3-Gal-9pathway with thelevel of IFN-γã€IL-10and IL-17.ã€Methods】Enzyme-linked immunosorbent assay (ELISA) was applied to detected theexpression of IFN-γã€IL-10and IL-17in peripheral blood plasma of preeclampsia andnormal pregnant women, and then a statistical analysis was needed.ã€Results】1.The level of IFN-γ and IL-17in preeclampsia patients were significantly higherthan that in normal pregnant women(P<0.05) and the level of IL-10inpreeclampsia patients was significantly lower than that in normal pregnantwomen(P<0.05).2.In preeclampsia patients, the IFN-γã€IL-17expression were positively correlatedwith the level of Tim-3and Gal-9protein, and the IL-10expression werenegatively correlated with the level of Tim-3and Gal-9protein.ã€Conclusion】IFN-γã€IL-10and IL-17were expressed in the peripheral blood plasma both ofthe preeclampsia patients and normal pregnant women. When the abnormalexpression of Tim-3-Gal-9pathway that can lead to the balance between Th1/Th2/Th17cells is disrupted, it may be related to the occurrence of preeclampsia. | | Keywords/Search Tags: | preeclampsia, maternal-fetal interface, decidua tissues, Tim-3, Gal-9preeclampsia, IFN-γ, IL-10, IL-17 | PDF Full Text Request | Related items |
