Objective:Our study was aimed to investigate the expression level of proteins related to BMP2signal pathway in kidneys of genetic hypercalciuric stone-forming rats and its effects on kidney interstitial calcification and urolithiasis.Methods:In this study,eighteen clean and three-months old male genetic hypercalciuric stone-forming rat(GHS) rats who were250g to300g, and eighteen similar male normal control(NC)SD rats were selected respectively. Fresh kidney tissues were obtained immediately after executed them.Von-Kossa calcium stain was used to detect calcification,Real time quantitative PCR and Western blotting were used to detect the mRNA and protein expression level of proteins related to BMP2signal pathway including BMP2,cbfal and Osterix in renal tissues.Statistical analysis was performed using the SPSS15.0,and experimental data were expressed as means±tandard deviation.Statistical comparisons were tested by T test,and P<0.05was considered statistically significant.Results:BMP2,cbfα1and Osterix of GHS rats were higher than NC rats in protein level respectively(0.21±0.06and0.13±0.06,0.39±0.20and0.22±0.09,0.24±0.04and0.15±0.06,P <0.05).The similar results were detected in mRNA expression level,BMP2,cbfα1and Osterix of GHS rats were5.21times,2.07times and11.21times higher than NC rats respectively,there was significant difference between two groups(P<0.05).There existed apparent renal interstitial calcification in GHS rats.Conclusions:Our study demonstrated hypercalciuria may activate BMP2signal pathway,and the proteins related to the pathway would participate in the regulation of urine calcium reabsorption.The up-regulation of these proteins is possibly involved in the pathopoiesis of idiopathic hypercalciuria. There existed apparent renal interstitial calcification in GHS rats.BMP2,cbfα1and Osterix may play a role in the renal interstitial calcification and urolithiasis.
