The Induction Of Mouse IPS Cells With Insulin-like Growth Factor-1and Exogenous Transcription Factors

Objective: To determine the effects of insulin-like growth factor-1on nuclearreprogramming.Methods: Use fiber cell medium containing insulin-like growth factor-1to culturemouse fibroblasts（MEF） for three days; under the same condition,use fibercell medium without containing insulin-like growth factor-1to culture MEFfor threedays as a negative control; Finally, infected MEF with retroviruses expressingOct4, Sox2, Klf4, and c-Myc. Evaluate the formed iPS clones.Results: The iPS cells were successfully obtained using insulin-like growth factor-1.on the eighteenth day after infection.The induction efficiency is about0.05%,whichwas nearly five times.more efficiently than the control group.Morover, APstaining,SSEA-1andOct4staining of the experimental group were positive.Conclusion: Insulin-like growth factor-1play an important role in the reprogrammingof iPS cells,it can improve the efficiency of four-factors induction system.
