Objectives To investigate the effect of Lutein on proliferation and apoptosis of human Tenon’s capsule fibroblasts in vitro.Methods The CCK-8method was used to assay the activity of human Tenon’s capsule fibroblasts at12h,24h,48h after treated with5,10,20,40ug/ml Lutein in vitro.The cultured fibroblasts were treated with Lutein in various concentrations(5,10,20,40ug/ml) for24hours,the Flow Cytometry method was used to evaluate the cell cycles and the Hoechst33258staining was used to measure the apoptosis of the cells.Results The proliferation of human Tenon’s capsule fibroblast at12h,24h,48h after treated with5,10,20,40ug/ml Lutein was inhibited significantly compared with the control and it was dose-dependent and time-dependent(P<0.05);the cell population was significantly increased at the G0/G1phase and decreased at S phase in a dose dependent manner after the cultured fibroblast were treated with in various concentrations of Lutein(5,10,20,40ug/ml)(P<0.05); the Hoechst33258staining showed that with Lutein treatment varying from concentrations of5ug/ml to40ug/ml,the rate of apoptosis increased observably(P<0.05).Conclusions Lutein could inhibit the proliferation of human Tenon’s capsule fibroblasts, Lutein also could stimulate the apoptosis of the cell.These findinds suggest that Lutein as a possible new drug for inhibition of filtering bleb scarring may be worthy of further investigation.
