Aim:To investigate the role which plasma DNA play in the thrombosis of DVT patient and the changes of concentration with the course of thrombosis.Method:Use the magnetic beads kit to extract the DNA from the plasma sample of DVT patient,quantitify them by real-time PCR.We used an established mouse model of DVT induced by flow restriction in the inferior vena cava (IVC) to study the relation between the concentration of plasma DNA and the course of thrombosis.Result:Obviously increment of plasma DNA was observed in the DVT patient,among which the patient with cancer was higest in the concentration of plasma DNA,and the patient with no common incentive showed unexpected higher concentration of plasma DNA.Also we found that the thrombus increased the concentration of plasma DNA only during the first24h after the thrombus formed.Conclusion:The plasma DNA may contribute in the thrombosis of DVT patient with no common cause through an unknown mechanism. By studying the mouse model,it suggested that the plasma DNA influence the thrombosis mainly in the initial period of thrombosis and soon be digested by DNase when thrombus stablized.
