Research In RHD Genotyping Method For RhD-negative Individuals And Preliminary Study On Molecular Characteristics Of Rhd-negative Individuals With Anti-D

[objective] The objective of our study is to observe a simple genotyping method for HanChinese and to study the molecular characteristics of RhD-negative individuals with Anti-Dpreliminarily，which can provide a preliminary theoretical basis for rational transfusion inRhD-negative patients and whether to conduct RhD immunoglobulin intervention forRhD-negative maternal with Anti-D.[Method]1）Through multiple PCR-SSP method the specific exon7and intron4of RHD）were detected of all specimens.2）Then promoter and exon10of RHD were detected fornegative samples further.3）Specific exon3,4,5,6,7,9of RHD were amplificated for allpositive samples of exon7,intron4, promoter or exon10detected.4）For positivespecimens of exon7and intron4，Del (K409K) were detected by the BAGeneRHD-TYPE Asia kit.5）The specimens including K409K were validated through thePCR-SSP method.[Results]1) In116cases,83cases were of the RHD deletion, accounting for71.5%, and8 cases were of partial deletion of RHD exons, accounting for6.9%.Del (K409K) weredetected in24cases, accounting for20.7%.2) RHD were deleted of23RhD negativeindividuals with anti-D detected.3) In27specimens with non Anti-D,10cases wereDEL,2cases were partial deletion of RHD exons, and15cases were deletion of RHD.[Conclusion]1)The genotyping method in our study was easy and could quicklygenotype for the RhD negative specimens.2) Deletion of the RHD gene may be the mainmolecular biological characteristics of RhD-negative individuals.3)DEL may be thesecond main features.4）Whether individuals with DEL could receive RhD-positive unit orRhD-positive pregnant women with DEL need anti-D immunoglobulin prevention wouldneed to research further.
