Purpose: Lymphoma is a group of malignant tumors with the characteristic of hugeheterogeneity because of it has different biological and clinical features. Of course itis a crucial event to provide correct diagnose and appropriate classification forclincians. Classical cytogenetic studies play a critical role in the diagnosis ofmalignant lymphomas. But until now they are much less widely used in clinicalcourse due to the difficulty of the chromosome cultivation.Methods: In our studies, we set up a optimize method of chromosome cultivationusing lymph node, spleen specimens or other tissue biopsy specimens from thepatients with suspected lymphoma. Besides we perform G-banded karyotyping andfluorescent in situ hybridization (FISH) on these34specimens.Results:30of these specimens have been successful karyotyped including20lymphomas,2solid cancers,8lymphoproliferative disorders. The histopathologyclassification of these20lymphomas is mentioned as follows. There are6diffuselarge B-cell lymphoma(DLBCL),followed by5follicular lymphoma(FL),1mantlecell lymphoma,1marginal zone lymphoma,3T-cell lymphoblastic lymphoma,2peripheral T-cell (PTCL),2angioimmunoblastic T cell lymphoma.85%of theselymphoma patients have clonal chromosomal abnormalities. The current numeraryabnormalities include number2,4,7,8,12,16,21chromosome and the deletion of sexchromosome. The recurrent structural abnormalities include1p,6q,11q,14q,17q.Besides we detect3kinds of distinctive abnormal karyotype.Conclusions: With these results, it is much better to discover the recurrentchromosomal translocations. The association of chromosomal karyotype analysis withpathology, immunology, genetics make great efforts to the correct classification oflymphoma and clear their diagnosis. Besides it is helpful for us to understand the relationship between the karyotype and mechanism of the non-Hodgkin lymphomas.
