Objective:This study was designed to approach the expression of Smurf1andBMP-2in osteosarcoma and the correlation between the two withimmounohistochemistry. In this study, RT-PCR in osteosarcoma from gene level.Clinical pathological data was analyzed associated with the expression level of Smurf1protein and gene for the correlation. And also probed into the clinical significance ofSmurf1upon incidence and development of osteosarcoma.Method:120cases of bone tumor specimens for operations from December2009toDecember2011were collected from XieHe Hospital of Huazhong University ofScience and Technology.36cases of specimens were from benign lesion of bone whilethe other84cases from osteosarcoma. The protein expression of Smurf1and BMP-2was tested with immunohistochemistry. RT-PCR method was used to test theexpression level of Smurf1mRNA. The x2test was used to test the relationshipbetween immonohistochemistry、RT-PCR results and clinicopathology characteristicsSpermancorrelation analysis method was used in correlation analysis. The differencewas statistically significant when P<0.05.Results1. Positive expression rate of Smurf1in osteosarcoma was (0.786±0.214).Positiveexpression rate of Smurf1in benign lesions of bone was (0.583±0.183).The differencewas statistically significant(x2=17.603,P<0.05).2. Positive expression rate of BMP-2in osteosarcoma was (103.57±13.23).Positive expression rate of BMP-2in benign lesions of bone was (152.19±19.42).Thedifference was statistically significant(x2=12.39,P<0.01).3.There was a positive correlation between Smurf1protein and BMP-2priotein(r=—0.598,P<0.05).4.The expression rate of Smurf1mRNA in osteosarcoma is obviously higer thenin benign lesions of bone. The difference was statistically signiticant(t=14.52, P=0.001)5. There is no correlation between expression of Smurf1protein or mRNA withsex, age, sites of metastatic tumor, and whether primary malignancy exists(P>0.05),but was relevant to differentiation degree of cancer cells(P<0.05). With decrease ofdifferentiation of cancer cells, positive expression rate of Smurf1protein and mRNAincreased.Conclusion1. The Smurf1expression rate in osteosarcomas is significantly higher than inbenign lesions of bone..2. There is a negative correlation between expression of Smurf1and BMP-2. Itsuggests that Smurf1may advance the formation and development of osteosarcomasby restrain.BMP-2.3. In the tissues of osteosarcoma, positive expression rate of Smurf1protein andmRNA increases with decrease of differentiation of cancer cells, but has no correlationwith sex, age, sites of osteosarcomas..Expression of Smurf1protein and mRNA isclosely related to the differentiation degree of cancer cells, Smurf1may become anindex for predicting potential of osteosarcomas.
