Objective: To study the significance and clinical value of expressions of a-smooth muscle actin (a-SMA) and transforming growth factor-beta (TGF-β1) in osteosarcomas.Methods: a-SMA and TGF-β1 were detected for the specimens of 36 osteosarcomas and 20 osteochondromas and 21 osteosarcomas (before and after chemotherapy in the same patient) by immunohistochemical Envision method and SPSS 11.5 software was used to analyze the significance and relationships of their expressions.Results: The positive rate of a-SMA was47.20 percent in osteosarcomas group and 60.70 percent in high-grade osteosarcomas group with a significant difference . The positive rate of a-SMA was zero in osteochondromas group and in low-grade osteosarcomas group with a significant difference(x2=13.56, P < 0. 01) . The positive expression rate of TGF-81 was41.70 % in osteosarcomas group and 51.30% in high-grade osteosarcomas group with a significant difference . The positive expression rate of TGF-β1 was zero in osteochondromas group and in low-grade osteosarcomas group with a significant difference (x2=11.38, P < 0. 01) . In osteosarcomas group , the expression of a-SMA was positively correlated with the expression of TGF-81 ( rs = 0.546, P < 0. 01) . In osteosarcomas group with chemotherapy , the expression of α -SMA and TGF- β 1 was negatively correlated with the expression before chemotherapy. TGF- β1 (p>0.05) , α-SMA(p>0.05). Fisher's Exact Test showed that patients with lung metastasis and recurrence had significant TGF- β 1and α -SMA expression rates than those with disease-free survival, α -SMA (p<0.05) TGF-β 1(p<0.05).in high-grade osteosarcomas group positive rates of a-SMA and TGF-β1 were significant difference than those in low-grade osteosarcomas group. ( α -SMA, Hc =15.3607, p<0.01), ( TGF- β1 Hc =16.8771p<0.001)Conclusion: High expression of a-SMA and TGF-β1 are closely correlated with invasion and lung metastasis of osteosarcomas.
