Objectives To study therapeutic benefit and possible mechanism of intravenous administration of bone marrow mesenchymal stem cells after spinal cord injury in adult rats .Methods 1.Rat bone marrow mesenchymal stem cells (rMSCs) from adult rats were cultured in vitro,their specific cellular surface marker were analysed with flow cytometer.2.Rats were subjected to a weight driven implant injury. There are 12 in the non-injuried group and 120 rats in the injuried group. Then the injuried group was randomly divided into transplantation group and control group.There were 7 time points in each group,n=6 in each point(n=12 in 3 day,7day,14 day).rMSCs or PBS were infused into tail vein at 10 minute s after injury.3.Functional outcome measurements using the Basso-Beattie-Bresnehan Score were performed 24 hours post-transplantation and 3 days and 7 days and 14 days post-transplantation.4.Light microscopy and electron microscopy were used to identify the morpholo-gy of the sections of injured spinal cord tissue 14 days post-transplantation.Immunohistochemical staining and laser scanning microscopy were used to identif-y rMSCs and expression of BDNF,NGF,TNF-α,IL-1βprotein in the injured spinal cord sections 3 days and 7 days post-transplantation.5. RT-PCR was used to measured expression of BDNF,NGF,TNF-α,IL-1βmRNA in the injured spinal cord sections 3 hours,6 hours,12 hours,24 hours,3 days,7 days,14 days post-transplantation.Results 1.
