Objective:This experiment using different concentrations of Bupleurum decoction of the role of in vitro cultured human hepatoma HepG2cell lines, To observe cancer cell survival under the drug action, inhibition rates of growth curve and vascular endothelial growth factor (vascular of endothelial growth factor, VEGF) and expression The different perspectives of the Bupleurum tumor suppressor mechanism and the best concentration of drug therapy.Methods:Human hepatoma HepG2cells were divided into control group (high glucose DMEM medium containing10%fetal calf serum),5-fluorouracil (5-Fluorouracil,5-FU)125ug/mL drug group, Bupleurum decoction10.5mg/mL,7mg/mL,3.5mg/mL different concentrations of the experimental group. Application thiophene MTT assay (MTT) test different concentrations Bupleurum decoction and5-fluorouracil role, the inhibition rate of cell viability and drugs on cell lines. Application of growth curve of different concentrations of drugs on human hepatoma HepG2cell line proliferation. Immunohistochemical staining of different concentrations of drugs on human hepatoma HepG2cells VEGF protein expression.Results:MTT results detect blank control group,5-Fu drug group, Bupleurum decoction10.5mg/mL,7mg/mL3.5mg/mL group of HepG2cell viability were1±0.089,0.244±0.035,0.483±0.038,0.88±0.044,0.991±0.086;5-Fu drug group, Bupleurum decoction10.5mg/mL,7mg/mL with blank control group, the difference was statistically significant (P <0.01or<0.05), Bupleurumdecoction the3.5mg/mL group with blank control group, no significant difference (P>0.05); of Bupleurum group with5-FU drug group difference was statistically significant (P<0.01), each Bupleurum groupdifference between statistical significance (P<0.01);5-FU drug group Bupleurum decoction10.5mg/mL,7mg/mL,3.5mg/mL, group inhibition rates were75.6%,51.71%,11.98%,0.89%; growth curve shows the5-Fu drug group, Bupleurum decoction10.5mg/mL,7mg/mL,3.5mg/mL group can inhibit cancer cell growth, Bupleurum decoction10.5mg/mL7mg/mL group inhibition Bupleurum decoction 3.5mg/mL group, with5-Fu drug group growth curve; immunohistochemical assay blank control group, Bupleurum decoction10.5mg/mL,7mg/mL,3.5mg/mL of VEGF expression of the integral value of3.64±0.265,1.48±0.160,2.48±0.204,3.2±0.126, the Bupleurum group, with the blank control group difference was statistically significant (P<0.01),The Bupleurum group difference was statistically significant (P<0.01).Conclusion:Bupleurum decoction certain amount of concentration can cause destruction of HepG2cells, and inhibit the growth of HepG2cells, and a dose effect relationship; Bupleurum decoction downward to the HepG2cells the expression of VEGF in a concentrationeffect relationship; Bupleurum decoction may have an effect on liver cancer treatment by a sufficient amount of the active ingredient.
