Tetrastigma obtectum (Wall.) Planch is the whole plant of Tetrastigma obtectum widely distributed in south of China. It has been used as traditional Chinese medicine for the treatment of pain in rheumatoid arthritis, traumatic injury, osteomyelitis, irregular menstruation and so on. However, no chemical investigation has been reported from this plant. For the report from the same genus, some flavones, steroids, organic acids have reported from related species.This article according to the research on traditional medicine of Tujia ethnic minority in hubei province, in order to search for new bioactive constituents from natural source, During the extaction and isolation experiment, including silica gel, toyopearl HW-40, gel permeation chromatography, and preparative HPLC,11compounds were isolated from95%EtOH extract of T. obtectum. By using UV、IR、 MS、1H-NMR、13C-NMR、2DNMR(1H-1H COSY, HMBC,HSQC, ROESY) spectra analysis and chemical reaction, the structures of isolated compounds were elucidated as follow:two indole alkaloids, tetrastigmindole A (1), tetrastigmindole B (2), eight flavones:cis-apigenin-6-vinyl-7"-rhamnoside (3), cis-apigenin-8-vinyl-7"-rhamnoside (4), apigenin-8-C-[6-deoxy-2-O-(α-L-rhamnopyranosyl)-xylo-hexopyranos-3-uloside](5), apigenin-8-C-[α-L-rhamnopyranosyl(1'2)]-rhamnopyranoside](6), apigenin-8-C-[α-L-rhamnopyranosyl(1'2)-xyloside](7), apigenin-7-O-β-D-glucopyranoside (8), luteolin-7-O-β-D-glucopyranoside (9), vitexin-α-L-rhamnoside (10), apigenin-6-C-α-L-arabinopyranoside (11). Compound1-7were new compounds.In the anti-tumor metastasis screening experiment of the indole alkaloids, tetrastigmanine A (1) and B (2) revealed positive effects on anti-tumor metastasis in MDA-MB-231cell lines at the concentration of20μg/ml. The value of inhibition ratio on MDA-MB-231is70.3%.In the experiment for GLUT4translocation in skeletal muscle L6cells, compounds1,3and5enhanced the GLUT4translocation, and revealed the activity of promoting the absorption of glucose.
