ObjectiveTo observe the hydroxyapatite-chitosan with Lysostaphin biomimetic materials in repairing rabbit radius bone defect,to provide theoretical basis and experimental basis for further research and clinical application of artificial bone material,to make a new treatment of bone defect and nonunion of bone.Methods36healthy adult New Zealand white rabbits were randomly divided into three groups of12rats were A Group:blank group;B Group:autogenous bone graft group;C group:material graft group.The middle of the Rabbit Radius with the periosteal bone segment resection of12mm long,made of standard bone defect model. A group do not make any disposement;B group are implanted autogenous bone;C group are implanted material.4,8,12weeks after theoperation,respectively for grosso bservation, X-ray plain film and histological observation of new bone repair materials,effects of bone repair.Results1.The general observations indicate that the defects of B group and C group have been the integration of shaping near normal at12weeks. The A group showed fracture line closed,do not form a bony connection.2. X-ray plain film shows that with the prolonged,B group and C group an increase in callus bone defect,12weeks of basic healing,shaping complete,A group no bone repair,the formation of nonunion. By Lane-Sandhu,X-film score P value:A group and C group compared with P<0.05significant difference;A group and B group than,P <0.05,indicates significant difference;B group and C group than in,P>0.05,no significant difference.3. Histological observation in each time period,in the fibrous connective tissue, callus,bone trabeculae and class as well as the amount of bone formation of osteoblasts in B group and C group were better than the A group. By Lane-Sandhu France histological score P value:A group and C group compared with P<0.05 significant difference; A group and B group than,P<0.05,indicates significant difference;B group and C group than in,P>0.05,no significant difference.Conclusions1.The hydroxyapatite-chitosan with Lysostaphin biomimetic composite materials has good biocompatibility,no toxicity and foreign body reaction,wound healing is normal,no wound infect,material expose and tissue necrosis after implant. The motioneating, mental state is good.2.The hydroxyapatite-chitosan with Lysostaphin biomimetic composite materials has good conductivity,provide support for new bone,there are much new bone tissue between material and bone after implant.3.The hydroxyapatite-chitosan with Lysostaphin biomimetic composite materials has good degradability,with the time passed,the material is degrading,and replaced by bone tissue, and this process almost synchronism.4.The hydroxyapatite-chitosan with Lysostaphin biomimetic composite materials has good bone formation, can be used as a new type of composite artificial bone to repair bone defects, and provide a new way for clinical treatment.
