Objective:To observe the new type of nano‐hydroxyapatite / chitosan‐ phosphorylated polyelectrolyte complexes of chitosan / gelatin / rhBMP‐2 biomimetic nano‐composite materials in repairing bone defect with the effect of bone tissue engineering scaffold material for further research and clinical application of theoretical basis and experimental basis for the bone defect and bone are not Even the treatment of new ideas.Methods:36 healthy adult New Zealand white rabbits were randomly divided into three groups of 12 rats were A: experimental group; B Group: autogenous bone graft control group; C groups: control group. The middle of the Rabbit Radius with the periosteal bone segment resection of 1.5cm long, made of standard bone defect model, the experimental group implanted Task Force developed nano‐hydroxyapatite / chitosan‐ phosphorylated chitosan polyelectrolyte complex / Gelatin / rhBMP‐2 biomimetic nano‐composite bone repair materials, autogenous bone graft autogenous bone implanted in the control group control, blank control group were not implanted in any material. 2,4,8,12 weeks after the operation, respectively for gross observation, X‐ray plain film and histological observation of new bone repair materials, effects of bone repair. Results:The general observations indicate that the experimental group and the autogenous bone graft group after 8 weeks after implantation, defects has been the integration of shaping near normal at 12 weeks. The control group showed fracture line closed, do not form a bony connection. X‐ray plain film shows that with the prolonged, A group and B group an increase in callus bone defect, 12 weeks of basic healing, shaping complete, C group no bone repair, the formation of nonunion. By Lane‐Sandhu, X‐film score P value: A group and C group compared with P <0.05 significant difference; B group and C group than, P <0.05, indicates significant difference; A group and B group than in, P> 0.05, no significant difference. Histological observation in each time period, in the fibrous connective tissue, bone trabeculae and class as well as the amount of bone formation of osteoblasts in A group and B group were better than the C group. By Lane‐Sandhu France histological score P value: A group and C group compared with P <0.05 significant difference; B group and C group than, P <0.05, indicates significant difference; A group and B group than in, P> 0.05, no significant difference.Conclusion:The nano‐hydroxyapatite / chitosan‐ phosphorylated chitosan polyelectrolyte complex / gelatin / rhBMP‐2 biomimetic nano‐composite material has good bone formation, can be used as a new type of composite artificial bone to repair bone defects.
