ObjectiveIn this study, A miniature pig experimental periapical periodontitis model was established, and the deciduous teeth pulp stem cell sheet induced by VitminC(Vc) was used to treat the bone defect, to study the osteogenic capacity of cell sheet, actually providing a support for clinical application of cell sheet.Methods:1Established miniature pig experimental periapical periodontitis model. Three miniature pigs were anesthetized, mandibular deciduous molar(Dm) crown occlusal access was made, the pulp was removed and the apical foramen was destroyed by#30K-files, the root canal was irrigated with saline solution. Then the root canals, dried by sterile paper points, were filled with10mg/ml LPS and the teeth were sealed with cement.2Miniature pig deciduous teeth pulp stem cells were cultured in60mm dishes and the cell sheet were induced by Vc(20μg/mL).3.1Experimental group A:deciduous teeth pulp stem cell sheet+β-TCP control group B:β-TCP blank group C:nothing3.2Miniature pig experimental periapical periodontitis was treated with the transplantation of deciduous teeth pulp stem cell sheet. Three miniature pigs were anesthetized and buccal mucous membrane of operating field was dissected, periapical periodontitis bone defect was filled with cell sheet+β-TCP composition.After this, mucosal flap was resetted and sutured.Three miniature pigs were killed after the transplantation of cell sheet12weeks.Results1Cell sheet was induced by VitaminC successfully.2The root canal was filled with LPS,4weeks later, the destruction of periapical bone was present on CT, to demonstrate that the experimental periapical periodontitis model was successfully established.3The periapical bone defect was cured by transplantation of cell sheet+TCP composition after12weeks.Conclusions:In this study, we successfully established the experimental periapical periodontitis bone defect model, and then cured the bone defect with transplantation of cell sheet+TCP composition, proving the capability and advantage of cell sheet. The development of Vc-mediated cell sheets may provide the basis of animal experiments for treatment of periapical periodontitis with stem cells.
