Objective: To investigate the effect of PRP/PDLSCs/HA in periodontal defects ofminiature swine.Methods: Collect the Guizhou miniature swine’ s venous blood and prepare thecentrifuged PRP two times. Porcine PDLSCs were harvested by using outgrowth method;Immunofluorescence was used to detect the expression of STRO-1of porcinePDLSCs.The third passage cells were cultured in three-dimension scaffold of HA, and thebiological characteristics of the cells were observed through the scanning electronmicroscope at the1st,3rd and7th day. Establish model of the miniature pig periodontalbone defects: Animals were randomly divided into three groups according to the materialtransplanted into bone defect site. Group A was added with PRP/PDLSCs/HA, group Bwith HA, and group C without any treatment except the suture of surrounding soft tissues.The progress of osteogenes is in each group was respectively observed in4,8and12wafter transplantation.Results: The cultured miniature swine PDLSCs shaped in typical fibroblast cells underlight microscope, Porcine PDLSCs expressed STRO-1, and the PDLSCs grew very wellon the scaffold of HA. Variouw amount of new bone formation appeared in bone defectsgrafted by group A and B, and group A was better than group B, but in blank control bonetrabeculae were scanty.Conclusion: The miniature swine PDLSCs, which can be cultured successfully in vitroand in HA, combination of PRP with PDLSCs in HA could enhance the bone healing considerably, this experiment can provide some basic theory for the regeneration of toothand periodontal tissues.
