| ObjectivesThis research tries to observe and compare, through applying acupoints medicine embedding method, the effects of collagen tetramethylpyrazine catgut, original collagen tetramethylpyrazine medicated thread and ligustrazine medicated thread on brain tissue MMP-9and TIMP-1of rats with focal cerebral ischemia reperfusion injury.MethodsTo some extent, the effects of embedding medicine and methods differ with deforming of medicated thread. As a result, this article undertakes research on production of ligustrazine medicated thread (because it is sol at less than37degrees, but become a semisolid gel at or above37degrees, so calling it the transformation of ligustrazine medicated thread), effects on brain tissue protein and also other factors in terms of safety and absorption rate ofmedicine.1. In terms of medicine safety research, healthy guinea pigs are chosen and a certain quantity of ligustrazine medicated thread shall be spread on the surface skin of pigs. Hypersusceptibi lity experiment shall be conducted later;0.5ml ligustrazine medicated thread shall be injected into muscle of healthy SD rats, then keep on observing for15days for any symptom like erythema, dropsy, asthma and shock.2. In terms absorption rate research, healthy SD rats shall be chosen and divided randomly into three groups namely collagen tetrameth-ylpyrazine catgut group, original collagen tetramethylpyrazine medicated thread group and ligustrazine medicated thread group.2mm length collagen tetramethylpyrazine medicated thread and collagen tetramethylpyrazine catgut and0.1ml same dosage ligustrazine medicated thread into shall be injected into Da Zhui and Nei Guan. These rats shall be visually observed after3,5and7days to check the absorption rate of medicine.3. In terms of effects research on brain tissue MMP-9and TIMP-1of rats with focal cerebral ischemia reperfusion injury,40nos. healthy SD rats with weight around200g shall be randomly divided into four groups namely model group, col lagen tetramethylpyrazine catgut group, collagen tetramethylpyrazine medicated thread group and ligustraz-ine medicated thread group. Each group has10nos. rats. Through modified thread embolism method experimental cerebral ischemia reperfusion model shall be made. Medicine shall be embedded in Da Zhui and Nei Guan. After24hrs all experimental rats ’heads shall be cut off and preserved in4%paraformaldehyde solution for6to8hrs, then embedding with paraffin. Immunohistochemistry method shall be used to detect the expression of brain tissue MMP-9and TIMP-1. HPIAS-1000computer high-resolution color image analyzer on slice image analysis, the results were analyzed statistically.Results1. Hypersusceptibility experiment shows that ligustrazine medicated thread does not lead to any symptom like erythema, dropsy, asthma and shock. 2. Absorption rate experiment show that three kinds of medicated thread still exist after3and5days. ligustrazine medicated thread becomes stable semisolid gelatin, collagen tetramethylpyrazine catgut and collagen tetramethylpyrazine medicated thread insocul-ates with tissue. After7days ligustrazine medicated thread still exists but turns out be less stable. The rest two medicated thread are almost absorbed.3. Immunohistochemistry experiment shows that masculine cells of brain tissue MMP-9increase greatly. Mascul ine cells of brain tissue TIMP-1stay less changed. There is great difference for expression of MMP-9and TIMP-1through comparing three groups with model group. Thereinto, the effect of ligustrazine medicated thread appears superior than other two groups (P<0.05); There is no big difference found when comparing collagen tetramethylpyrazine catgut and collagen tetramethylpyrazine medicated thread groups (P>0.05)。Conclusions1. ligustrazine medicated thread can be a safe and slowly absorbed new medicated thread in the medical practice.2. ligustrazine medicated thread can increase the level of TIMP-1and decrease MMP-9so that degradation of vascular basement membrane will be reduced. Then the integrity of brain vessel can be maintained to improved cerebral ischemia. The effect of ligustrazine medicated thread is excelled collagen tetramethyr-azine catgut and collagen tetramethyrazine medicated thread. It is closely related to the character of the slow absorption. |
