Liposarcoma is a malignant tumor composed of fat cells of different differentiation and atypia. It is one of the most common tumor of soft tissue in the adult, taking the20%of the sarcoma of soft tissue. Each position of the whole body can occur, but see mainly after deep soft tissue of lower limb and peritonaeum. It divides into on histology well differentiated, dedifferentiated, myxoid, round cell, and pleomorphic five shape types liposarcoma, and according to its cell and molecular biology feature divide it into3big kinds:well differentiated/dedifferentiated liposarcoma, myxoid/round cell liposarcoma and pleomorphic liposarcoma. Well differentiated liposarcoma accounts for40-45%and is easy to recur but not shift, low malignant level can make progress to dedifferentiated liposarcoma. Surgery remains the cornerstone of treatment for liposarcoma, but for patients those with disease that cannot be completely resected or with metastasis, novel targeted agents is urgently needed. ZIC1(Zic family member1), which was cloned and identified in1994, is one of five ZIC family genes and notable for its involvement in multiple developmental processes. Recent studies have shown that ZIC1plays an important role in many tumors.In the current study, we investigated ZIC1expression in adipose tissue tumor using IHC and ISH. In light of the research reports that ZIC1can adjust cell proliferation and apoptosis by regulating some cell cycle proteins, we also detect the expressions of cyclinD1, p16and Ki67and analyze their relationship between ZIC1and clinical significance in liposarcoma.Materials and Methods1Materials:Test to collect52cases, including12WDLPS,18MLPS,20DDLPS and2PLS. Additionally, to collect20lipoma,15normal fat tissue and10GIST(Gastrointestinal stromal tumor).2Methods:MaxVision immunohistochemical staining methods and in situ hybridization were used to detect the expression of ZIC1in liposarcoma and control group. MaxVision immunohistochemical staining methods were used to detect the protein expression of cyclinD1, p16and Ki67.Results1The overexpression rates of ZIC1mRNA and protein were80.8%(42/52) and76.9%(40/52),and the control groups were all negative or weak positive. The expression pattern between LPS and other control groups had statistical difference(P<0.01).2The overexpression rates of cyclinD1, p16and Ki67were67.3%(35/52),80.8%(42/52) and51.9%(27/52),which was much higher than that of lipoma and normal fat tissue(P<0.05).3The overexpression of ZIC1and cyclinD1in LPS was obviously associated with invasion or metastasis(P<0.05), but not with tumor sites(P>0.05), differentiation degree (P>0.05) and primary or recurrence group (P>0.05). No relationship was found between p16overexpression and clinicopathological features. The overexpression of Ki67was obviously associated with differentiation degree(P<0.05), but not with tumor sites(P>0.05), invasion or metastasis(P>0.05) and primary or recurrence group (P>0.05). Overexpression of ZIC1was positively correlated with high expression of cyclinD1in LPS(P<0.05), but no statistical relationship with p16or Ki67was observed(P>0.05).Conclusion Overexpression of ZIC1, cyclinDl and p16in LPS may play an important role in the carcinogenesis and progression of LPS and maybe helpful in the differential diagnosis.
