1. Abstract: Objective:To study the change of vascular smooth musclereactivity in T2DM rats and intervention of Ber, then find out therelationship between vascular smooth muscle reactivity and BKCachannel and the possibilem echanism of Ber in curing diabetic and itscomplications.Methods: feeding with high-fat and high-glucose foodand then injecting a low-dose of STZ(25mg kg) to establish type2diabetic rats model. All rats were divided into three groups randomly:the normal control group, the model group and the Ber group. TheBer group was orally administered with Ber (125mg/kg),rats in normalcontrol group and model group were orally administered with water.4weeks later, the all rats were killed one after another. Then observe thereactivity to KCl,PE and the relaxation of vascular rings induced bySNP before and after adding IbTX. Use Chart6.0software to analysisdata. Results:1Modeling:Model group: compared with the normalcontrol group,FBG, FINs, blood lipid, blood pressure,water intake, urineoutput,and food intake increased significantly however the weightreduced significantly.2.The relaxation percentage induced by SNP(10-7,5×10-7,10-6,5×10-6and10-5mol∕L)(1) Normal control group(n=16):before adding IbTX:13.87±4.98,51.81±12.37,81.68±14.91,90.83± 26.18,102.79±20.54; After adding IbTX:9.58±3.41,42.30±11.27,69.36±10.54,82.54±16.45,91.51±16.43,There were nosignificant differences (P>0.05).(2) Model group(n=16): before addingIbTX:18.75±4.23,53.49±11.49,82.65±14.86,102.93±28.13,104.99±20.13;After adding IbTX:6.58±2.61,30.24±7.94,47.36±11.73,73.54±16.19,81.42±13.49, There were significant differences(P<0.01).(3)Ber group(n=16): before adding IbTX:18.17±4.62,64.93±14.37,93.15±16.88,109.50±21.22,112.99±23.89;After addingIbTX:4.42±2.74,18.50±7.93,40.01±11.21,56.17±9.35,61.57±15.31,There were significant differences (P<0.01).(4) The difference ofthe relaxation percentage of thoracic aorta rings between before andafter adding IBTX in the three groups:Normal control group:7.20±2.12,9.14±4.01,12.40±3.54,8.09±3.28,8.90±3.75;Model group:11.47±2.02,27.54±11.31,35.29±9.51,30.43±7.81,31.27±8.73,compared with normal control group,it increasedsignificantly(except10-7mol L,P<0.01);Ber group:14.32±5.21,48.53±15.03,54.26±10.74,53.62±14.43,55.37±12.58,compared withmodel group,it increased significantly(except10-7mol L,P<0.05).4.Therelaxation percentage of mesenteric artery vascular rings induced bySNP(10-7,5×10-7,10-6,5×10-6and10-5mol∕L)(1)Normal control group(n=16): before adding IbTX:14.53±5.18,32.23±10.16,50.27±18.25,60.43±12.47,66.37±15.14; After adding IbTX:7.23±2.72,25.98 ±6.93,42.95±11.32,54.93±19.73,69.90±14.34,There were nosignificant differences (P>0.05).(2) Model group(n=18): before addingIbTX:19.69±4.98,39.48±10.73,60.35±14.98,69.41±17.65,72.37±15.47;After adding IbTX:4.37±1.32,8.54±3.21,20.97±7.39,23.61±6.89,25.20±8.12,There were significant differences(P<0.01).(3)Bergroup(n=16): before adding IbTX:23.50±8.73,60.22±20.11,85.12±11.14,87.19±10.29,88.01±19.57; After adding IbTX:6.18±2.85,11.79±5.29,24.11±5.02,31.44±10.74,34.43±9.29,There were significantdifferences(P<0.01)。(4) The difference of the relaxation percentage ofmesenteric artery vascular rings between before and after adding IBTXin the three groups:Normal control group:8.20±3.42,9.60±4.02,12.40±4.57,11.01±2.98,10.90±3.75;Model group:12.47±2.32,33.97±11.47,39.29±9.81,40.29±12.11,40.23±7.92,compared with normalcontrol group,it increased significantly (except10-7mol L,P<0.01);Bergroup:15.54±8.04,54.76±13.91,64.56±14.79,61.63±13.36,61.17±12.55, compared with model group,it increased significantly(except10-7mol L, P<0.05). Conclusion:1. The T2DM rat model wasestablished successfully.2. The reactivity of thoracic aorta andmesenteric artery to SNP were enhanced with the intervention of Ber.3.After adding IbTX,the change of the reactivity of thoracic aorta andmesenteric artery to SNP is different: Ber group>model group>normalcontrol group.
