Objective:To observe the effects of calcium sensitizers MCI-154 on vascular reactivity following normal rat; we also observed the effects of calcium sensitizers MCI-154 on artherosclerosis(AS) vascular reactivity through establishing AS model of rat to provide information for further clinical practice.Methods:Forty rats were divided into two groups randomly :normal group and model group,and each group was further divided into two subset groups: MCI-154 group and no MCI-154 group. Normal rats feed foundation animal feeds;AS rats feed hypso-lipin animal feeds to establishing AS model. The vascular ring of normal rats and AS rats was from thoracic aorta,and which was put in a constant temperature isolated organ perfusion system. To observe the effect of MCI-154 on different accumulative total concentration norepinephrine (NE) inducing contraction of the vascular ring. Tension/ humid weight of unit tissue (g/mg) was deemed to quantification standard;through it we drawed the quantity-effect curve, and evaluated vascular reactivity with average contraction force and the quantity-effect curve.Result:1. The influence of MCI-154 on normal vascular reactivity:â‘ the influence of MCI-154 on proper angiotasis of normal group: without NE inducing contractile response,each concentration MCI-154 can directly diminish the average angiotasis.â‘¡the influence of MCI-154 on normal angiotasis with NE inducing contractile response: MCI-154 can obviously diminish the average angiotasis with each concentration NE inducing contractile response,shift the quantity-effect curve to right, and offer dose dependent. With same concentration NE inducing contractile response, there were great difference between MCI-154 of each concentration and no MCI-154(P<0.01)or(P<0.05);as well there was great difference among all concentration (P<0.01) or(P<0.05).2. The influence of MCI-154 on AS vascular reactivity:â‘ the influence of MCI-154 on proper AS angiotasis: each concentration of MCI-154 can directly degrade the reactivity of AS vascular, and depressing extent differently arrive 1.1%,9.8%,10.1%,10.4% of the same concentration normal group. without NE inducing contractile response, MCI-154 can still directly diminish the average angiotasis ,but the group of 10-7mol/L has no obvious change.Furthermore,there is difference among all dose group of MCI-154 (P<0.05).â‘¡the influence of MCI-154 on AS angiotasis with NE inducing contractile response:AS vascular reactivity to contractile response induced by NE was significant depression, especially when concentration of NE arrived 10-7mol/L(P<0.01),without MCI-154 the average angiotasis drop to 55.8% of the same concentration of normal group; the quantity-effect curve significantly shifted to right. MCI-154 further diminished AS vascular reactivity,as well offered dose dependent. But among all dose group comparison, there was difference only between 10-4mol/L group and 10-7mol/L group(P<0.01)or(P<0.05).Conclusion :MCI-154 pretreatment decreased the reactivity of normal vascular ring.With regard to the decreasing reactivity of AS vascular ring, MCI-154 pretreatment can further decrease this vascular reactivity.
