Objective: Osteoarthritis (osteoarthritis, OA) is common inthe elderly prone to disease, progressive degeneration of articular cartilagedamage and osteophyte formation pathological features, cartilage, IL-1β inthe OA disease process plays an importantrole. IL-1β, IL-1familymember of the IL-1β receptor complex formed by combination of cartilageon the cell membrane IL-1RI and IL-1R accessory protein (IL-1RAcP) andIL-1RI and IL-1RAcP in the cell domain dimerization between differentsecond messenger (cAMP, GTP) information delivery system, the activationof IL-1signal transduction. In normal cartilage and normal synovial tissue ofIL-1β amount is small, but had higher levels of expression in OA cartilage.The acupuncture treatment through the stimulation of points around the knee,and promote the body’s release of endogenous morphinetie and otheranalgesic substances, pain threshold, to achieve the purpose of analgesia.Electro-acupuncture therapy can make muscles a Shu received, played therole of the pump, thereby improving the local blood circulation to promotethrough the gas run, absorb exudate, and is conducive to the restoration ofthe articular surface of cartilage. This subject by observing theelectro-acupuncture treatment of the cartilage of rabbit knee OA grossmorphological changes, pathological changes of chondrocytes, and cartilageof IL-1β expression impact study electrical effect and mechanism of chondrocyte repair of rabbit knee OA model. Methods:(1) Objects andinterventions: new zealand white rabbits (male)36, random sample of12normal control group, no modeling, and the remaining24rabbits wererandomly divided into12of12of the model group and EA group. The obtained numerical results using thex±s, P <0.05wasconsidered significant difference, P <0.01significant difference. Results:(1)Cartilage is generally observed: normal control group left knee was nosignificant joint effusion, the appearance of smooth cartilage surface waswhite orchid, deep and bright color, no crack, the cartilage structure meanmankin ’s’s score of0. Most of the model group left knee joint effusion, allthe cartilage surface greyish yellow, rough articular surface, some parts ofthe erosion and gloss lower surface of the cartilage surface is slightly uneven,rough; most of the animals of the EA group joint effusion and synovialhyperplasia, cartilage change mainly to the rough, but to a lesser extent;electro-acupuncture treatment group and model group’s score according tomean Mankin ’s cartilage structure, there is significant difference (P <0.05).(2) HE staining and toluidine blue staining pathology observed: normalcontrol group, HE staining shows cartilage surface roughness, the cartilagelayer of uniform thickness; chondrocytes arranged in neat rows, cell size isconsistent and complete, the tide line, stained with toluidine blue uniform,no matrix loss of staining; model group, HE staining showed cartilagesurface ulceration missing cartilage thickness was significantly thinner, asignificant reduction in chondrocyte cells gather in cluster, disorders of thetide line, toluidine blue staining was significantly diminished; EA group cartilage surface compared to the rules, fissures, cartilage cells arranged inneat, compared with model group, the cartilage cells were significantlyincreased thickening of the cartilage thickness, showing the disorder of thetide line, the mild decline of toluidine blue staining. The normal controlgroup and model group, the mean Mankin ’s’s score between the modelgroup and EA group, the differences were significant (P <0.05).(3)Cartilage immunohistochemistry of IL-1β measurement conditions: normalcontrol group, only surface occasionally IL-1β weakly positive expression inchondrocytes, little brown intracytoplasmic granules; the cartilage matrix ofthe model group, the cytoplasm in both IL-1β expression, the whole layercan be seen, especially in the middle the highest density of mature cartilagecells more dark brown brown particles; EA treatment group, present in theIL-1β expression in the cartilage surface can be seen scattered in thecytoplasm of chondrocytes, light brown brown granules. The normal controlgroup and model group, model group and EA group comparison, thedifferences were significant (P <0.01). Conclusion:(1) The production ofstraight plaster cast fixation rabbit knee OA model can simulate OA diseaseprocess modeling success of this experiment.(2) IL-1β promote the role ofthe articular cartilage extracellular matrix (extracellular matrix, ECM)degradation and increased cartilage damage, the damage is very important.(3) Electro-acupuncture is effective in treating knee OA, and its mechanismmay be associated with reduced cartilage content of IL-1β, promote the synthesis of proteoglycans, the repair of cartilage cells, prompting theincrease in the content of cartilage matrix, etc. have a certain relationship.
