Objective:The effect and mechanism of action of Hedyotis diffusa to asthma mice airway inflammation and remodeling.Methods:1. Preparations for the models of the asthma mouse40Male BALB/c mice, weight of18-22g, were randomly divided into4groups,and the groups were asthma group, contra] group, Hedyotis Diffusa high-dose group(high-dose group), Hedyotis Diffusa low-dose group (low-dose group). Except contral group other mice were sensitized on the first day and14th day by intraperitoneal injection of20μg OVA emulsified in1mg of aluminum hydroxide in a total volume of200μl. On the21st,22nd, and23rd day after the initial sensitization, the mice were challenged for30min with an aerosol of3%OVA in saline (or with saline alone as a control). As for the observation, restlessness, abdominal muscle twitching, shortness of breath in the mice were regarded as the positive reaction. Except the control group and the normal group, the other two groups were treated with intragastric administration of appropriate doses of drugs from the day before inhalation. After inhalation, all groups of mice were executed and the serum and lung tissue were collected by experimental design.2. The ELISA was used to observe the number and classification of the cell in the BALF.3. PAS and HE staining and transmission light microscope were used to show the changes of pulmonary structure and the effects on inflammatory infiltration and structural damages in asthma lung tissue after inhibition of RAS activation and chymase activity.4. The western blot was performed to detect the expression of NF-kB in the right lung tissues.Results:In asthma group, the number of inflammatory cells and the expression of IL-4,IL-5,IL-13in BALF is higher than the control group, IFN-y were lower; NF-kB in lung is significantly higher than those in control group (P<0.05). Hedyotis Diffusa significantly decreased. The number of inflammatory cells, the expression of IL-4.IL-5.IL-13in BALF, the content of NF-kB in lung tissues were significantly lower. but IFN-γ were higher, with differences (P<0.05). For index mentioned above, there were no significant differences between Hedyotis Diffusa high-dose group and Hedyotis Diffusa low-dose group (P>0.05).Conclusion:The Hedyotis Diffusa inhabits the level of IL-4, IL-5, IL-13, NF-kB and increase the level of IFN-γ in asthmatic model to inhibition asthma attack and airway inf-lammation.
