Objective: To study the effect of the expression of Glycogen synthase kinase-3β(GSK-3β) and the content of hepatic glycogen in the liver and muscle glycogen in the muscle of mice with Diabetes mellitus (DM) by Dragon's blood (Sanguis Draconis).Method: Selecting 120 mice of Kunming strain, and randomly dividing them into six groups-normal group, DM group, C4H11N5·HCL group and Dragon's blood groups with three dosage (125mg/kg·d,250mg/kg·d,500 mg/kg·d). The C4H11N5·HCL group were given C4H11N5·HCL(100mg/kg·d) by introgastric (ig) everyday for 7d. Dragon's blood groups were given Dragon's blood by introgastric (ig) everyday for 7d. The indices were observed and recorded. For example, determining the weight and the Fasting plasma glucose(FPG) of DM mice, the content of hepatic glycogen in the liver and muscle glycogen in the muscle of mice with DM, the expression of GSK-3βin the liver and right-back leg' s muscle of mice with DM by Immunocytochemical and Western blot(WB).Result: After given Dragon's blood,it can be shown from the statistics that the weight of DM mice rose and the FPG of DM mice decreased (P<0.05) After given Dragon's blood, the content of hepatic glycogen in the liver and muscle glycogen in the muscle of mice with DM can be increased (P<0.05), and the expression of GSK-3βin the liver and right-back leg's muscle of mice with DM can be decreased (P<0.05)Conclusion: Dragon's blood could increase the content of hepatic glycogen and muscle glycogen in the liver and muscle of mice with DM. Dragon's blood could inhibit the expression of GSK-3βin the liver and muscle of mice with DM. I could infer from the statistics, tables and picturesthat Dragon's blood can decrease Plasma glucose by decreasing GSK-3β.
