Objective:To investigate the effect and mechanism of myocardial hypertrophy and fibrosis of Spontaneously hypertensive rats by observing the effect of Xuefuzhuyu capsule on blood pressure, left ventricular mass index, left ventricular function, pathological change at myocardium, collagen content, MMP-9、 TIMP-1in Spontaneously hypertensive rat.Methods:Thirty-two male spontaneously hypertensive rats aged forteen weeks were randomly and equally into four groups:high dosage group(treated with Xuefuzhuyu capsule of8g/kg-d), low dosage group(treated with Xuefuzhuyu capsule of4g/kg-d), Luotingxin group(1mg/kg-d), model group, The age-matched Wistar-Kyoto rats(WKY)were used as control. Measure systolic blood pressure every two weeks, After forteen-week-treatments, calculated the left ventricular mass index and left ventricular function, HE and Masson staining was used to observe the pathological changes of myocardium tissues, The ultrastructural changes of myocardial cells were observed by electron microscopy, the changes in the expression and content of Ⅰ,Ⅲ collagen and MMP-9, TIMP-1protein were detected with Immunohistochemistry.Results:Compared with control group, systolic blood pressure and left ventricular mass index were increased while systolic function of left ventricle were decreased in the model group, pathological change obviously at myocardium stained with HE,Masson trichrome and electron microscopy, Increased expression level of Ⅰ, Ⅲ collagen and MMP-9, TIMP-1, compared with the model group, the ratio of LVW/BW is decreased of Xuefuzhuyu capsule, the fibrosis were closer to the luotingxin group,the pathological change of myocardium is alleviative, Type Ⅰ,Ⅲ collagen was determined.Conclusions:Xuefuzhuyu capsule can alleviate myocardial hypertrophy and fibrosis induced in spontaneous hypertensive rat, the mechanism may be related to regulate the expression of collagen Ⅰ, collagen Ⅲ, MMP-9and TIMP-1.
