ObjectiveTo investigate the effect of sulforaphane (SFN) on human small cell lung cancer cell line NCI-H446invasion and matrix metalloproteinase (MMP)-9expression, and then try to provide reference for exploringeffective drugs for lung cancer.MethodsNCI-H446cells were cultured in vitro, then treated with OμM,25μM,50μM,100μM of SFN for24-72h. Cell proliferation and its ability to adhere to fibronectin were observed by MTT assay; Migration and invasion of NCI-H446cells treated by SFN were detected by Transwell assay. Total cellular RNA was isolated, MMP-9mRNA level was detected by RT-PCR. MMP-9enzymic activity was tested by gel zymogram experiment.Results1. MTT assay showed that different concentrations of SFN (25μM,50μM,100μM) could inhibit NCI-H446cells growth in a dose-and time-dependence manner. After treated for72hs, the inhibit rate of was11.1%,25.2%and44.6%, respectively.2. The control cells without SFN could grow normally on the cell culture plate with extracellular matrix. In the condition of SFN, the cells shapes had changed immensely, and the adherent cells decline in the culture plate when the concentration reached up to100μM. MTT assay shows that the different concentrations of SFN could significantly decline the adherence of NCI-H446cells to extracellular matrix, the suppression rates increased with the ever-increasing concentrations of SFN. 3. SFN could inhibit NCI-H446cells invasion, and the effects enhanced with the ever-increasing concentrations of SFN. When25μM,50μM,100μM of SFN treated for24h, the amount of invading cell per field in cell invasion assay were (85.4±4.41)%,(68.8±6.84)%and (50.5±4.26)%, respectively.4. RT-PCR showed that various concentrations of SFN could decrease the MMP-9mRNA level in a dose-dependent manner in NCI-H446cells.5. Gel zymogram experiment showed that the activity of MMP-9in NCI-H446cells was declined significantly with the treatment of SFN.Conclusion1. SFN could inhibit NCI-H446cells proliferation, and inhibit them adherent to the extracellular matrix.2. SFN could also inhibit CNI-H446cells migration and invasion.3. The inhibitory effect of migration and invasion might be relation to the declined expression of MMP-9and the downregulated enzyme activity.
