Study On The Effect Of Lycopene On Hepatic Lipid’s And Brain Lipid’s Metabolism In Hyperlipidemia Rats

OBJECTIVE The aim of this experiment is to study the regulation of lycopene on the metabolism of lipid in the liver and brain in hyperlipidemic rats, provide evidence for further exploitation and application of lycopene.METHODS According to the weight and serum TC level,40health male SD rats which fed nomal diet for1week were divided in to5groups:group A with nomal diet, group B with diet contain0.5%propylthiouracil, group C with high fat diet, group D with high fat diet and lycopene, group E with high fat diet and fluvastatin, the experiment will last for3weeks. From the second week to the end, group D feed40mg·kg·bw-1·d-1lycopene directly into the stomach. group E feed10mg·kg·bw-1·d-1fluvastatin directly into the stomach. After the animal expriment, some indicator which related to lipid metabolism in serum, liver and brain will be tested, contain total cholesterol(TC), triglycerides (TG), high density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C), total antioxidant capacity(T-AOC), apolipoprotein E(ApoE), low density lipoprotein receptor(LDLR),24-hydroxyl cholesterol(24HyC).RESULTS1. Feeding high fat diet for1week cause hyperlipidemia;rat’s serum and hepatic TC,TG,LDL-C level increased significantly and HDL-C level decreased; Feeding high fat diet can raise rat’s brain TG and lower down brain HDL-C level,but it has no effect on brain TC,LDL-C,T-AOC level,the number of nervous cells in hippocampus CA1decreased, and the nucleus has pyknosis, parts of the cytoplasm broken and most of the cells denaturated.Feeding diet contain0.5%propylthiouracil can raise serum and hepatic TC,TG,LDL-C level significantly and decrease HDL-C level. Feeding diet contain0.5%propylthiouracil can decrease brain HDL-C level in rats,but it has no effect on brain TC,TG,LDL-C.the number of the cells in hippocampus CA1decreased, all the cells are clearly arraged, the nucleus are round and parts of them has pyknosis.2. Gavage with lycopene or fluvastatin can significantly reduce serum and hepatic TC, TG, LDL-C level, and raise serum and hepatic T-AOC level in hyperlipidemic rats; Lycopene and fluvastatin can reduce brain HDL-C level and has no effect on brain TG level.After the gavage with lycopene or fluvastatin for3weeks,rat’s brain ApoE,LDLR and serum,hepatic24HyC level increase, serum and hepatic ApoE level decrease; serum,hepatic LDLR and brain24HyC level didn’t change. The rats gavage with lycopene,the cells in hippocampus decreased, parts of the cytoplasm broken and there are pyknosis; gavage with fluvastatin has the similar effects with lycopene except more physaliphore.3. Hepatic TC,TG,LDL-C and serum TC,TG,LDL-C has positive linear regression, and with negative linear regression to serum HDL-C; Negative linear regression exists between hepatic HDL-C and serum TC,TG,LDL-C.Brain TG has positive linear regression with hepatic LDLC,serum and hepatic LDLR,24HyC and brain ApoE,LDLR,24HyC, brain TG has negative linear regression with serum HDL-C and ApoE and hepatic HDL-C.Brain HDLC has positive linear regression with serum HDL-C and ApoE and hepatica HDL-C and ApoE,brain HDL-C has negative linear regression with serum TC and24HyC,hepaticTC,LDL-C and brain LDLR,24HyC.Serum T-AOC has positive linear regression with brain TG, and has negative linear regression with serum TG,brain LDL-C. Hepatic T-AOC has no linear regression with serum lipid,hepatic lipid,brain lipid.Brain T-AOC has positive linear regression with hepatic TG and brain LDL-C.CONCLUSION1. Gavage with40mg·kg·bw-1·d-1lycopene in hyperlipidemia rats,serum and hepatic TC,TG,LDL-C level decreased,hepatic HDL-C and brain TG level increased. Serum T-AOC level increased. Lycopene has protection to brain hippocampus CA1cells, indicate that brain TG might be correlation with the protection.2. Lycopene can intervene hepatic TC,TG,HDL-C,LDL-C level through the regulation in serum TC,TG,HDL-C,LDL-C level. and affect brain TG level through the regulation in serum24HyC, brain LDLR,24HyC level. and affect brain HDL-C level through the regulation in serum, hepatic TC,TG,LDL-C leve, serum LDLR,24HyC, hepatic24HyC, brain LDLR,24HyC level.3. Hyperlipidemia was induced in rats after feeding high fat diet for1week; after3weeks, rat’s serum, hepatic TC,TG,LDL-C and brain TG incereased, brain HDL-C level in rats, there is no effect in brain TC,LDL-C level, and the nervous cells in hippocampus CA1changed pathologically. Feeding diet contain0.5%propylthiouracil in rats, serum and hepatic TC,TG,LDL-C increased and brain HDL-C level decreased, brain TC,TG,LDL-C level didn’t change, hippocampus CA1nervous cells pathologically changed, indicate that lycopene might be used to intervene these changing in rats feeding with0.5%propylthiouracil.
