Analysis Of ORMDL3Polymorphisms And Its Association With Serum OPN And TGF-β1Level In Childhood Asthma Of Hunan Province

Objective:To investigate the distribution characteristics of single nucleotide polymorphisms of ORMDL3gene in Han Chinese children of Hunan Province and analysis the relationship between ORMDL3SNPs and atopic status and asthma susceptibility.To approach the correlation of ORMDL3SNPs with serum OPN and TGF-β1levels.Methods:98cases were0-5years old children with asthma as asthma group and30healthy children were same age as control group,who came from Hunan Provincial Children’s Hospital since December2010to October2011.On family members within3generations with or without allergic diseases asthma group was divided into two groups,with atopic and non-atopic group.The SNPs of rs7216389of ORMDL3gene were detected by direct DNA sequencing.Serum OPN and TGF-β1level were detected by enzyme linked immunosorbent assay. The relationships of ORMDL3gene polymorphism and serum OPN and TGF-β1level were analyzed by using correlation analysis.Results:(1)Analysis of ORMDL3gene polymorphism:There were three genotypes AA,AG and GG in the rs7216389site of ORMDL3.The frequency of AA,AG and GG genotypes of rs7216389were respectively68.4%,23.5%and8.1%in asthma group,and60.0%,26.7%and13.3%in control group.The allele frequency of A and G were respectively80.1% and19.9%in asthma group,and73.3%and26.7%in control group. There was not significantly difference between the two groups about genotype and allele frequency(χ2=0.994and1.248,P>0.05for all).The frequency of AA,AG and GG genotypes of rs7216389were respectively71.0%,22.6%and6.4%in asthma with atopic group,and63.9%,25.0%and11.1%in with non-atopic group.The allele frequency of A and G were respectively82.3%and17.7%in atopic group,and76.4%and23.6%in non-atopic group.There was not significantly difference between the two groups about genotype and allele frequency(χ2=0.829and0.984,P>0.05for all).(2) Compare of serum OPN,TGF-β1content:The contents of serum OPN were respectively18.68±4.94ng/mL and13.53±3.55ng/mL in asthma group and control group.Compared with the control group the contents of serum OPN was significantly increased in asthma group (t=5.296,P<0.05).The contents of serum OPN were respectively18.71±5.53ng/mL and18.63±3.81ng/mL in atopic group and non-atopic group.Compared with the control group the contents of serum OPN was significantly increased in atopic group and non-atopic group (P<0.05).There was no significant difference of the OPN content between the atopic group and non-atopic group and between each Genotype of all the groups(P>0.05).The contents of serum TGF-β1were respectively69.86±31.76pg/mL and47.06±23.76pg/mL in asthma group and control group.Compared with the control group the contents of serum TGF-β1 was significantly increased in asthma group(t=3.630,P<0.05).The contents of serum TGF-β1were respectively79.03±32.96pg/mL and54.08±22.29pg/mL in atopic group and non-atopic group.Compared with the control group and non-atopic group the contents of serum TGF-β1was significantly increased in atopic group(P<0.05).There was no significant difference of the serum TGF-β1content between the non-atopic group and control group and between each Genotype of all the groups(P>0.05).(3) Correlation analysis:The serum OPN and TGF-β1contents were liner positive correlation in asthma group, atopic group and non-atopic group.Conclusions:(1)There was single nucleotide polymorphisms of rs7216389site of ORMDL3gene in Han children of China’s Hunan Province.There may be no significant relation between SNPs of ORMDL3gene and asthma susceptibility in Han Chinese children of China’s Hunan Province.(2) The SNPs of rs7216389site of ORMDL3gene may be not the SNP loci affecting the atopic of asthma children of Han Chines in China’s Hunan Province.(3) OPN and TGF-β1may involve in the occurrence and development of asthma,and there was a linear positive correlation between them.(4)The SNPs of rs7216389site of ORMDL3gene did not affect the expression of OPN and TGF-β1.
