Objective The mechanisms of oxymatrine on rat hepatic stellate cell lines were elucidated through the following three aspects. First, we studied on the inhibition of oxymatrine on HSC-T6proliferation. Second, we observe the effect of oxymatrine on HSC-T6activation in vitro.Thirdly, expressions of telomerases was studied at different time.Methods HSC-T6cell lines were incubated and divided into the control group and three treated groups (2mg/mL,4mg/mL and6mg/mL respectively), The expression of cell proliferation, tissue inhibitor of Metalloproteinase-1(TIMP-1), α-smooth muscle actin, types Ⅰ(Col Ⅰ mRNA) and Ⅲ (Col ⅢmRNA) collagens, and rTERTmRNA by reverse transcriptase polymerase chain reaction technology.Results Proliferation of HSC-T6cell lines was dependent on the concentration of oxymatrine and incubation time. Inhibition effect of oxymatrine was strengthened along with the elevated concentrations concentration and prolonged incubation time. The difference of expression of α-SMAmRNA,Col Ⅰ mRNA,Col ⅢmRNA and TIMP-1mRNA between the control group and treated groups were not statistically significant after24hours (p>0.05).Incubated for48h,6mg/mL oxymatrine promoted the expression of α-SMAmRNA and TIMP-1mRNA, the difference was statistically significant (P<0.05), while others were not. Incubated for72hours, compared with the control group, expression of Col Ⅰ mRNA, Col ⅢmRNA and rTERTmRNA of each group were not significantly different (P>0.05),but4mg/mL and6mg/mL oxymatrine groups got significantly lower expression of α-SMA and TIMP-1mRNA than the control group, difference was statistically significant P<0.05).Conclusions Firstly, the effects of oxymatrine on proliferation of hepatic stellate cells were dependent on concentration of oxymatrine and incubation time. Secondly, the oxymatrine was an effective inhibitor of activation of HSC by lower the expression of α-SMAmRNA and TIMP-1mRNA.Thirdly,oxymatrine was not effective for syntheses of Col Ⅰ and Col Ⅲ.Finally, oxymatrine stimulated the expression of telomerase at24hours, but on effects was observed when prolonged the incubation time.
