Camptothecin is a kind of plant anti-cancer drug, it attracts great interest due to its unique anti-cancer property. Chemosynthesis of camptothecin was regarded as complex and by-products accumulated. Also, directly extracting from plants is considered to harm both camptothecin’s bioactivity and the environment. Therefore, this paper aims to develop an environmental healthy way to obtain camptothecin through endophytic fungi fermentation.In this paper, camptothecin was found in the secondary metabolites of a strain of endophytic fungus named EFCI-13after fermentation cultivation detected by TLC and HPLC analysis. Microbial form identification showed that EFCI-13is categoried for gibberellin spores.Moreover, we optimized the culture medium of EFCI-13when considering the influence factors including fermentation time, culture temperature, and the addition amount of MgSO4. The experiment scheme was designed by Box-Benhnken method considering the relationship of each variables. The results shows that the best fermentation condition is that: fermentation time with7days, culture temperature with27℃, the addition amount of MgSO4with1.4g according to the software analysis to the surface figure and contour map of each factor.Further study focused on microbe transformation of EFCI-13to spices. Under optimized conditions, EFCI-13converts raisin extracts to spices through microbe transformation. Two important spices in the raisin extract:2,3-dihydro-3,5-dihydroxy-6-methyl-4H-Pyran-4-one and5-HMF were detected by GC-MS analysis during the whole process to determine their content changes. The results showed that the content of the two substances increased by3.0times and3.2times separately after biotransformation by the enzyme extracts of EFCI-13while increased by87.7times and52.3times after biotransformed derectly by active hyphae in growth period of EFCI-13compared with raw materials.
