Objective:To study pharmacodynamic relationship between HPLC fingerprint and anti-influenza virus in order to evaluate and control quality of Flos Lonicerae Japonicae, which ensures efficacy of clinical use of drugs, establishing spectrum-efficiency relationship mathematical models of anti-influenza virus for quality assessment method and basis.Method:Firstly, investigate the extraction method and chromatographic conditions and establish HPLC fingerprint of Flos Lonicerae Japonicae with the technology of fingerprint of TCM after collecting all the major producing areas of Flos Lonicerae Japonicae samples all over the country; secondly, investigate the efficacy of anti-influenza virus of Flos Lonicerae Japonicae samples using experimental methods of vitro anti-influenza virus and half of the therapeutic concentrations as the evaluation index; following analysis of experimental results by means of mathematical methods of multivariate polynomial fitting, association of fingerprint data and efficacy results, creation and subsequently verification of Spectrum-efficient mathematical model.Results:Collected of90batches of Flos Lonicerae Japonicae samples and totally found6characteristic peaks in common. Established a HPLC fingerprint of Flos Lonicerae Japonicae with more peaks, high resolution and good repeatability, Determined the efficacy results is EC50=0.261mg/mL-2.019mg/mL,TI=14.57-39.85. Established computer mathematical model of spectrum-efficiency relationship of the Flos Lonicerae Japonicae anti-influenza virus according to experimental data of80batches of Flos Lonicerae, using mathematical methods of multivariate polynomial fitting samples, which was verified by another10batches of Flos Lonicerae samples.Conclusion:The method that the efficacy results are judged by mathematical methods and the determination of the TCM chromatographic fingerprints is accurate and feasible. The institution of spectrum-efficient mathematical model helps the result of judgment more scientific, more comprehensive and more realistic. Because of good accuracy and feasibility, the method helps TCM quality assessment more intuitive, more rational and more effective, and also has great significance to the quality evaluation of Chinese herbal medicines and the safety of clinical drugs.
