| Objective:The experiment observe VCI rats with the Shouwuyizhi Capsule by cognitivefunction change situation and detect the expression change of VEGF-Notch/Delta signalpathway-related proteins,(VEGF,Notch-4,Dll-4) change and observe the behavior change.Find the theoretical and experimental basis for the clinical application of the ShouwuyizhiCapsule.Method:SD rats were used and the VCI model was established with MCAO.80rats werechosen from120rats by Morris water maze and randomly divided into sham operationgroup, model group, Piracetam group and Shouwuyizhi group. After4weeks’medication,the rats’behavioral change was detected by Morris water maze. VEGFexpression was measured by immunohistochemistry method and the expression of theprotein Notch-4and Dll-4were measured by Western blot.Results:(1).Morris water maze experiment showed that the rats, the escape latency and Numberof cross-platform of ShouWuYiZhi Capsule group was better than that of rats of modelgroup and Piracetam group,more approaching sham operation group. Comgpared with themodel group,the sham operation group,Piracetam group,ShouWuYiZhi Capsule group hasa significant difference(P<0.01); Comgpared with the sham operation group,Piracetamgroup has a difference(P<0.05), ShouWuYiZhi Capsule group has not the statisticallysignificant(P>0.05); Comgpared with the, ShouWuYiZhi Capsule group a difference(P <0.05).(2).In the protein expression of VEGF and Notch-4. Sham operation group, modelgroup, Piracetam group, ShouWuYiZhi Capsule group in turn increase. Comgpared withthe Sham operation group,the other groups have a significant difference(P<0.01);Comgpared with the model group,the Piracetam group, ShouWuYiZhi Capsule group havea significant difference(P<0.01); Comgpared with the Piracetam group, ShouWuYiZhiCapsule group has a difference(P<0.05).(3). In the protein expression of Dll-4: Sham operation group, model group, Piracetamgroup, ShouWuYiZhi Capsule group in turn increase. Comgpared with the Sham operationgroup,the other groups have a significant difference(P<0.01); Comgpared with the modelgroup,the Piracetam group, ShouWuYiZhi Capsule group have a significant difference(P<0.01); Comgpared with the Piracetam group, ShouWuYiZhi Capsule group has asignificant difference(P<0.01).Conclusion:ShouWuYiZhin Capsule regulate the protein VEGF,Notch-4,Dll-4in the pathwayVEGF-Notch/Delta signaling, by this way, promote the regeneration of brain tissueischemic lesions, blood vessels and surrounding tissue. Improve the oxygen supply andpromote the damaged neurons recovery, thus improve cognitive function effectively. |
