Cervical carcinoma is one of the most frequently malignant tumors in women over the world, which serverely threats the health and life of female patients. Recently its incidence and mortality increases significantly and has the tendency of juvenescence. Generally it takes about ten years from cervical intraepithelial neoplasia to cervical carcinoma. Therefore, it is significant to reduce the incidence of cervical carcinoma by discovering cervical intraepithelial neoplasia in early phase and obstructing it in the appropriate way.As an oxygen sensor, prolyl hydroxylase domain, oxygen-dependent catalyse hydroxylation of specific proline residues in the HIFs-alpha subunits, mediates the degradation of HIFs in protein expression and transcriptional activity. It induces the coding of vascular endothelial growth factor, erythropoietin, transferrin, insulinlike growth factors-a and so on, which participate in tumor growth, energy metabolism and angiogenesis. It can be seen clearly that PHD2plays an important role in the process of tumor formation and development. Prolyl hydroxylase domain is becoming a hotspot in recent years, however, it is rare in the study of gynecological tumor at home and broad. There is no relevant research reported about the research of PHD2in cervical diseases at home. It helps us to understand the role of PHD2in the process of tumor formation and development by studying the expression of PHD2in cervical diseases. And it will supply a new though to clinical medicine.Objective To investigate significance of PHD2in the development of cervical cancerization by detecting the expression of PHD2in normal cervix, cervical intraepithelial neoplasia and cervical carcinoma. Methods Immunohistochemistry method and reverse transcription polymerase chain was used to detect the expression of PHD2in normal cervix as experimental group, CIN Ⅰ and CINⅡ,CINⅢ, cervical carcinoma as control group, which were respectively20cases obtained from the Gynecology Department of the First Affiliated Hospital of Anhui Medical University.Results In normal cervix, CIN Ⅰ and CIN Ⅱ, CINⅢ, cervical carcinoma, the optical density ratio of PHD2mRNA/GAPDH were respectively0.366±0.034、0.486±0.040、0.619±0.076、0.706±0.107. The difference of four groups was significant (P<0.01). The mean density of PHD2were respectively0.155±0.040、0.235±0.043、0.322±0.027、0.369±0.022. The difference of four groups was significant (P<0.01).Conclusion (1)The level of PHD2had a increasing tendency in the development of cervical cancerization showing that PHD2plays an important role in the process of tumor formation and development.(2)The difference of normal cervix and cervical intraepithelial neoplasia Ⅰ-Ⅱ was significant. The expression of PHD2in in early stage of cervical disease is rising, showing that PHD2could diagnosis cervical disease in early phase as a new tumor marker.(3)The expression of PHD2is rising gradually along with more serious cervical disease. It has much clinical value of PHD2for the prognosis and follow-up of cervical disease.(4)The difference of cervical carcinoma and normal cervix was significant. The discovery of PHD2may offer a new therapeutic target of cervical carcinoma by downregulating the expression of PHD2.
