Objective:To establish a model of chronic abacterial prostatitis,CAP of SD-rat and toinvestigate expression and significance of transient receptor potentialM8(TRPM8)between model group and control group.Method: An antigen solution was made by mixing the SD rats, purified prostate proteinand the Freund’ s complete adjuvant(FCA).The model of CAP was established byinjecting the solution subcutaneous with a multi-point way. Every rats of the CAP groupwas injected0.5ml DPT througt intraperitoneal.Dissected prostate in aseptic conditionafter8weeks. Select the back lateral lobe in the clean bench and stripping the prostatecapsule, blood vessels, fat and other tissues.Part of tissue used for HE staining, Thebigger part store in Rnalater for RT-PCR.Trizol was used to extract total RNA ofTRPM8,Reverse Transcription and SYBR Green I Real Time RT-PCR was usedto measure mRNA expression of TRPM8.Result:The expressions of TRPM8was found both in CP group and control group, andthe expression of TRPM8in CP group was decreased in contrast to control group0.811±0.044、0.937±0.050(P<0.05). Conclusion:The Application of Freund’s complete adjuvant modeling method to buildrat chronic prostate model is similar to the performance of pathological findings andpathology results of clinical chronic prostatitis and it provide a basis for further study ofthe pathogenesis of chronic prostatitis.These results suggest that the number of TRPM8was decreased in CP, We speculate that it may involved in the mechanism of CP.
