Object to detect the promoter methylation status of RASSF1A in the tissue andperipheral blood of patients with laryngeal caricinoma, in order to investigate therelationship betweenthe promoter methylation of RASSF1A and laryngeal caricinoma.Methods methylation specific polymerase chain reaction(MSP) was used to detectpromoter methylationstatus of RASSF1A gene in laryngeal caricinoma tissues andadjacet tissues from52patients, and24specimens of normal were detected with thesame method as well.And, the relationship between the promoter methylation ofRASSF1A and the clinical and pathological features was analyed.At the same, thepromoter methylation status of RASSF1A gene in peripheral blood of35patients withlaryngeal caricinoma and24specimens of normal was detected to investigate therelationship between the promoter methylation of RASSF1A and laryngeal caricinoma..Results(1)The positivity rate of promoter methylation of RASSF1A gene wassignifucantly highter in laryngeal caricinoma than that in the adjacent tissues andnormal controls(65.4%、7.7%、0%p<0.05), but on significant difference was detectedbetween the adjacent tissues and normal contrals(2)It is found that the promotermethylation of RASSF1A gene had no correlation with age, gender, differentiationdegree, T stage (p>0.05)(3)The frequency of promoter methylation of RASSF1A genein peripheral blood of the patients with laryngeal caricinoma and the specimens ofnormal, were54.3%,0%, respectively, with significant difference among the(p<0.05)Conclutions (1) promoter methylation of RASSF1A genen is present in laryngeal caricinoma, suggesting it might be associated with the development of laryngealcaricinoma.(2) promoter methylation of RASSF1A gene had no correlation with thedifferentiation degree, T stage, suggesting it may provide help in the earlydevelopment of laryngeal caricinoma.(3) The detection of RASSF1A promtermethylation might be an important molecular marker for the earlier diagnosis oflaryngeal caricinoma. At the same time, MSP is a novel and specific method formethylation detection around the promoter region,which may used in diagnosis oflaryngeal caricinoma.
