Background:Oral squamous cell carcinima(OSCC) which account for80%of the oralmalignant tumors is the most common malignant tumor in oral and maxillofacial.Currently the main treatment is surgery excision, and the adjunctive treatment isradiotherapy and chemotherapy. Extracellular matrix metalloproteinaseinducer(EMMPRIN) is a member of the immunoglobulin super-family, which is existsin epithelial cells, germ cells, left ventride myocardial cells and vascular endothelialcells under normal circumstances. But it is highly expressed in tumor cells. It also canlead to tumor growth and invasion through a variety of ways. Matrixmetalloproteinase(MMPs) is a kind of connective tissue synthesis and secretion ofproteolytic enzymes family, and its major role is to degrade basement membrane andextracellular matrix, promoting tumor invasion and metastasis. Because of the lack ofprotein homologous sequences, MMP-7is the smallest member with the molecularweight, of the MMPs.Purpose:In this study, EMMPRIN and MMP-7expression in OSCC and normal oralmucosas(NOM) were detected by immunohistochemical SP techniques, and thedifferent expression of MMP-7mRNA between OSCC and NOM was detected byreverse transcription-polymerase chain reaction(RT-PCR). In order to explored theirrelationship between the occurrence and development of OSCC.Methods:In the study,62cases of OSCC were selected as the experimental group, and11cases of NOM were selected as the control group. We used immunohistochemical SPtechniques to detect the expression of EMMPRIN and MMP-7in OSCC and NOM, and analyzed the relationship with the clinical and pathological data. RT-PCR was usedto investigate the expression of MMP-7mRNA in18cases of the experimental groupand5cases of the control group.Result:1. The positive expression rate of EMMPRIN in OSCC was80.65%(50/62), higherthan that in NOM which was45%(5/11), and the difference of the rate is statisticallysignificant(P<0.05). The positive expression rate of MMP-7in OSCC was also80.65%(50/62), higher than that in NOM which was18%(2/11), and the difference ofthe rate is statistically significant(P<0.05).2. The expression of EMMPRIN in OSCC has positive correlation with tumor size,lymph node metastasis and clinical stage(P<0.05), but not correlated with patients’age and gender(P>0.05). The expression of MMP-7in OSCC has positive correlationwith lymph node metastasis and clinical stage(P<0.05), but did not correlate withpatients’ age, gender and tumor size(P>0.05).3. Positive relationship between EMMPRIN and MMP-7expression was observed inOSCC (γ=0.801).4. The positive expression rate of MMP-7mRNA was66.7%(12/18), but negativeexpression in NOM, and the differences of the rates are statistically significant(P<0.05).Conclusion:The abnormal of EMMPRIN and MMP-7is significantly associated with thedevelopment of OSCC, and both of them are participated in the invasion and metastsisof OSCC. The two factors may play a synergistic role in the development andprogression of OSCC.
