PURPUSE:With the study of the effect of recombinant human bone morphogenetic protein-2(rhBMP-2) and rabbit external maxillary vascular bundle on osteogenesis in the process of rabbit mandibular defects repairing with chitosan(CS) and β-tricalcium phosphate (β-TCP) cement, discuss the feasibility of the method. METHODS:Place CS/β-TCP cement with rhBMP-2into the rabbit mandibular defects, and embed external maxillary vascular bundle into scaffold materials. A total of36New Zealand rabbits were divided into experimental group with CS/β-TCP/rhBMP-2and maxillary vascular bundle involved (n=12),one control group without vascular bundle (n=12),and another control group without rhBMP-2(n=12).The rabbits were sacrificed at4th,8th and12th week after the operations. The mandibular specimens were assessed by gross observation, X-ray image, bone density value and hematoxylin-eosin staining observation. RESULTS:The bone density value of the experimental group was higher than that of the control groups at8th and12th week (P<0.05). At12th week, gross observation of the experimental group’s specimens showed that boundaries of the materials were difficult to identify by naked eyes; no obvious difference was observed between the specimens and autogenous bone in X-ray performance; hematoxylin-eosin staining showed rich mature trabecular bone and lamellar bone. The indices mentioned above were better than that of the control groups. CONCLUSION:rhBMP-2and external maxillary vascular bundle can work together to help osteogenesis in the process of rabbit mandibular defects repairing with CS/β-TCP cement.
