Objective: To analysis the effect of Cuscuta chinensis polysaccharide local injection combined with distraction osteogenesis in treatment of rabbit tibial bone defects. To identify that Cuscuta chinensis polysaccharide can accelerate the process of bone formation, and to preliminary clarify the mechanisms of traditional Chinese medicine in promoting osteogenesis.Methods: 24 mature New Zealand white rabbits weighing 2.5 ± 0.5 Kg were randomly divided into polysaccharide treatment group and saline injection control. The rabbits were anaesthetized with 3% barbitone sodium solution, and then fracture was made in their left tibial before they were fixed in the unilateral external fixator. 0.5 m L of normal saline were locally injected once after surgery and 1 week later respectively; in the experimental group an equal volume of Cuscuta chinensis polysaccharide(0.5ml:200mg) were locally injected at the same time. After surgery one week, two weeks, three weeks and six weeks, general situation observation, X-ray technique were applied to dynamically detected new bone mineralization; Micro-CT scanning and biomechanical measurement were used to evaluate the effect of distraction osteogenesis combined with Cuscuta chinensis polysaccharide local injection in treatment of rabbit tibial bone defects. In this process, its extent to bone formation promotion was evaluated, and bone formation difference between the groups was analyzed.Results: 1. The general situation and the X-ray examination: at day 24 after the distraction, data on X-ray and mineral density showed a significant difference in bone density percentage between bone defects group and the control group.2. Micro-CT bone scan:at week 6 post the stretch experiment, the treated group showed a larger amount of bone formation than the control group. And the density was significantly higher.3. Biomechanical test results showed that the maximum torque angle dodder experimental group, maximum torque and torsional stiffness were significantly higher than the saline control group(P < 0.05); while there was no significant difference in the deg values between the two groups(P > 0.05).Conclusion: Cuscuta chinensis polysaccharide injection promoted bone formation in the Ilizarov external fixator distraction for treatment of rabbit tibia bone defects; the bone mass was greater and ossification was more complete; Cuscuta chinensis polysaccharide local injection can accelerate osteogenesis and shorten the healing time, suggesting Cuscuta chinensis polysaccharide have a good practical value in fracture healing...
