Objective:To observe type2diabetes mellitus (T2DM) rats model with the extension of the course, AP which is correlated with the glucose and lipid metabolism indicators changes in rat blood and brain. Exploring the effect of berberine on Aβ and the possible mechanism of berberine.Methods:Wistar male rats feed with high-fat diet for8weeks and streptozotocin (STZ) to establish T2DM rat model. The experimental animals were divided into5groups according to blood glucose levels and the weight:control group (Ctrl),model group (Model), berberine high-dose group (BerH), low dose group the (BerL) and positive control pioglitazone group (PIO). All groups received intragastric administration daily for8weeks,one times per day.At the end of the day of the administration,The model rats were taken blood from retrobulbar venous plexus.To observe fasting glucose(FPG),glycosylated serum protein(GSP),triglycerides(TG), total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol (LDL-C) change by colorimetry,and insulin content by radioimmunoassay,and the serum advanced glycation end products levels,serum A(340and serum Aβ42levels by enzyme-linked immunosorbent assay.all rat brains were removed, the enzyme-linked immunosorbent assay measuring brain changes of Aβ40and Aβ42in brain.Results:1.Compared with the control group, after feeding with high-fat for five weeks, body weight of wistar rats was significantly increased (P<0.05),for6-8weeks,body weight was very significantly increased (P<0.01);after feeding with high-fat diet for8weeks, FPG, Insulin was significantly increased (P<0.01),insulin sensitivity index (IAI) was significantly decreased (P<0.01).Rats were induced Insulin resistance (IR).High fat diet after8weeks,rats were intraperitoneal injection STZ with35mg/kg,A criteria to T2DM motel is fasting blood glucose>16.7mmol/L.after72h.compared with control group,Serum glucose, triglycerides, cholesterol, low-density lipoprotein cholesterol were significantly increased (P<0.01), high-density lipoprotein was significantly decreased, but insulin did not change a little.rats have been successfully replicated T2DM model with the characteritic of insulin resistance and lipid disorders.2.To observe the correlation of Aβ in brain with glucose and lipid metabolism indicators after modeling T2DM rats for8weeks. Glucose and lipid metabolism were still abnormal.Compared with control group, Apin blood and in brain significantly increased in T2DM rat (P<0.01).The Aβin brain and Aβ in blood was highly negatively correlated (P<0.05),But no correlation with FPG, GSH, AGEs, INS, TG, HDL-C and LDL-C. The increase of Aβ in T2DM rats brain has nothing to do with the glucose and lipid metabolism, is mainly close to the Aβ of peripheral blood,The incidence of Aβ deposition in T2DM rat Aβ content change rate in brain accounted for56.6%.3.The possible mechanism of Berberine improve Aβ deposition:T2DM rats were administrated after the intervention,BerH,PIO significantly reduced the content of Aβ in rats brain and blood(P<0.01, P<0.05),BerL can reduce Aβ in the brain and blood but without statistical significance.Conclusion:High-fat diet for8weeks and STZ35mg/kg can successfully replicate T2DM rat model on the basis of IR and high glucose and fat;Aβ in brain and Aβ in blood are highly negative correlated in T2DM rats, but has nothing to do with the glucose and lipid metabolism indicators. Aβ content change rate in T2DM rats brain was50.0%.BerH can significantly reduce Aβ level in T2DM rats brain and blood.
