Objectives:To investigate the influence of hypoxia on the change of generation andinvasion ability in human colonic cancer cell line SW480and it’s relationship withthe expression of hypoxia-inducible factor-1α(HIF-1α) and Snail, cultured humancolonic cancer cell line SW480ovarian in vitro.Methods:1)The human colonic cancer cell line SW480were respectively exposed toCoCl2(100,200,400,600,800μmol/L),and proliferation were detected by MTTmethods in order to select suitable CoCl2concentration. According to the CoCl2concentration, human colonic cancer cell line SW480were respectively cultivated tonormoxia and hypoxia.(2)Cells migration and invasion were detected by Transwellmethods.(3) Immunocytochemistry and RT-PCR and Western blot were used todetect the change of the HIF-1α and Snail in the human colonic cancer cell lineSW480.Results:Hypoxia significantly decreased cell survival rate, and the vital impulse ofSW480decreased with the increase of the concentrations of CoCl2, it confirmed theclose relationship between the proliferation and survival with hypoxia, and the abilityof cell migration and invasion were enhanced in hypoxia; The results of RT-PCR andWestern blot were showed that mRNA and protein expression of HIF-1α and Snailwere high expressed in hypoxia, Similar results were observed inimmunocytochemistry, furthermore the expression of Snail were enhanced with thethe increase of HIF-1α.Conclusion:The ability of cell migration and invasion were enhanced in hypoxia;The over-expression of HIF-1α and Snail might be important biological marker formalignant transformation and invasion and metastasis of colon carcinoma.Snail maybepromote the expression of the HIF-1α;...
