Objective: By establishing severe acute pancreatitis(SAP) model inSprague-Dawley rats, we observe the pathology changes of pancreas andintestinal.The rats are treated by edaravone.we investigate the intervention effect ofedaravone injection and its mechanisms of protection on intestinal mucosa barrierfunction injury of SAP with the purpose of providing evidence for preventing andcuring intestinal mucosa barrier function injury of SAP in clinical practice.Methods:Fifty-four adult male Sprague-Dawley rats weighting300-340g wererandomly divided into three groups:control group(group C),severe acute pancreatitisgroup(group SAP) and treatment group with Edaravone(group T).Each group wasrandomly divided into three subgroups as the phases of3h、6h、12h afteroperation. Rats in group SAP were induced by the retrograde injection ofbilio-pancreatic duct with5%sodium taurocholate(1.0ml/kg),but rats in group C,5%sodium taurocholate was displaced by0.9%sodium chloride.In group T,Edaravone(3mg/kg) was injected from the rat tail vein at once after the severe acutepancreatitis(SAP) model was established. Rats were killed after observed onschedule.The dynamic changes of the indexes of intestinal mucosa barrier functioninjury and pancreas injury at various time points in each group,incluing the levels ofserum AMS、ET、MDA、MPO and TNF-α.Expression of ICAM-1were determinedby immunohistochemical technique.Results:1.In comparison with group C,the serum levels of AMS,TNF-α,ET weresignificantly increased in group SAP(P<0.05).Compared with group C,the serumlevels of AMS,TNF-α,ET were significantly increased in group T(P<0.05),butgroup T was short of group SAP.2.In comparison with group C,the levels ofMDA,MPO were increased in group SAP(P<0.05); Compared with group SAP,the levels of MDA,MPO were reduced in group T (P<0.05).3.Expression of ICAM-1inileum tissues in group C was almost not detected,but overexpression of ICAM-1could be observed in group SAP(P<0.05); Compared with group SAP,Expression ofICAM-1in ileum tissues in group T was reduced(P<0.05).4.The rats in group SAPintestinal histopathology napped part fracture, edema and thickening, ileal mucosallayer edema, hyperemia and submucosal capillary congestion; pathological changeswith pancreatic tissue hyperemia blood, necrosis, edema and inflammatory cellinfiltration; group C intestinal and pancreatic the histopathologic scores were notstatistically significant (P>0.05),SAP group of rat intestinal and pancreatic tissueinjury obviously, its pathological score higher than those of C group (P <0.05); groupT intestinal and pancreatic tissue pathology score was lower than that of SAP group(P <0.05), but still higher than that of group C.5.There was positive correlationbetween the serum level of ET and the serum level of MDA(r=0.836,P<0.05),thesame as the expression of MPOin ileum tissues (r=0.853,P<0.05)、the expression ofTNF-α(r=0.828,P<0.05).In addition to,the expression of ICAM-1in ileum waspositively correlated with the serum level of ET(r=0.872,P<0.05).Conclusions:1.The gut barrier in rats is damaged in the early phase ofSAP.2.The expression of ICAM-1in ileum tissues in rats is significantly enhanced inthe early phase of SAP.3.Edaravone can relieve gut barrier injury in experimentalSAP that we can be inferred that Edaravone can reduce oxygen free radicals,inhibitinflammatory reaction and the expression of endothelial cells ICAM-1....
