Objective: To observe the effect of caveolin-1on TNF-α, IL-6and IL-10expression during vascular anastomosis stenosis.Method: The model of vascular anastomosis was established by anasto-mosing the rabbit carotid artery.40rabbits were randomly divided into fourgroups: normal control group (false operation group), surgical group, blanktransfected group and Caveolin-1transfection group, which from the AnimalCenter of University of South China. Rabbits were executed7and14daysafter operation. Among this, the specimens of7days for real-time PCR andwestern blot, and the specimens of14days for HE staining and immunohisto-Chemistry. Hematoxylineosin staining was used to observe the changes of arter-ial intima, real-time PCR, western blot and immunohistochemical method wereused to detecte Caveolin-1, TNF-α, IL-6and IL-10expression of mRNA andprotein.Result:1. HE staining showed that the intimal of surgical group andblank transfected group proliferated significantly, and so the vessel’s wall wasthickened, while in caveolin-1transfected group the intimal proliferation andthe blood vessel’s thickening was not conspicuous.2. The results of real-timePCR showed that mRNA expression of TNF-a and IL-6in caveolin-1transfect-ed group were significantly lower than the surgical group and the blank trans-fected group, but higher compared with the control group (P<0.05). Expressionof IL-10mRNA in caveolin-1transfected group was higher than surgical group and blanktransfected group(P<0.05). Compared with normal group, caveolin-1mRNA expression of surgical group and blank transfected group was signifi-cantly lower (P<0.05), while the expression of caveolin-1mRNA in the Trans-fected group was significantly higher than the surgical group (P<0.05).3. Wes-tern blot analysis showed that caveolin-1protein expression in the surgical gr-oup and blank transfected group was significantly lower compared with thenormal group (P<0.05), while the protein expression of caveolin-1in transfect-ed group was significantly higher (P<0.05).Compared with normal group, TNF-a and IL-6protein expression in the surgical group and blank transfected gr-oup was significantly enhance (P<0.05), while the TNF-a and IL-6protein ex-pression in the transfected group was significantly decreased (P<0.05).Comparedwith normal group, IL-10protein expression in the surgical group and blanktransfected group was enhanced (P<0.05), IL-10protein expression in the trans-fected group was significantly increased than surgical group (P<0.05).4.Immu-nohistochemical staining showed that TNF-a and IL-6expression in the trans-fected group was significantly weakened, and IL-10expression in the surgicalgroup and blank transfected group was significantly enhanced compared withthe surgical group and blank transfected group.Conclusion:1. Caveolin-1can inhibit neointimal proliferation after carotid artery anastomoticoperation in the rabbit.2.Caveolin-1can regulate inflammatory response by inhibiting the TNF-α and IL-6expression, as well as up-regulating IL-10expression at vascular anastomosis.
