| Objective This study is to investigate the molecular mechanism of EGFRvâ…¢ promotingmigration and invasion of hepatocellular carcinoma cell.Methods The differentially expressed proteins were analyzed and verified betweenHuh-7-EGFR and Huh-7-EGFRvâ…¢ cells by comparative proteomics. S100A11and Cofilin-1were found be differentially expressed proteins.The complete CDS of S100A11or Cofilin-1were introduced into Huh-7cells using a lentiviral expression systems,in order to constructthe Huh-7cell strains which is overexpression of S100A11or Cofilin-1gene.Then, themigration and invasion of Huh-7-S100A11and Huh-7-Cofilin-1cells were analyzed andverified. Small interfering RNA(siRNA)targeting the mRNA of human interference theS100A11or Cofilin-1were constructed, these siRNAs with Lipofectamine were packaged andtrasfected the mixture into Huh-7-EGFRvâ…¢ cells.After that, the migration and invasion ofsiS100A11Huh-7-EGFRvâ…¢ and siCofilin-1Huh-7-EGFRvâ…¢ cells were analyzed andverified.At last, S100A11and Cofilin-1and EGFRvâ…¢ in20pairs of carcinoma and adjacenttissues of hepatocellular were detected by real-time quantitative PCR to analyz therelationship between S100A11and EGFRvâ…¢ or Cofilin-1and EGFRvâ…¢.Results Five kinds of differentially expressed protein were indentified by the comparativeproteomics between Huh-7-EGFR and Huh-7-EGFRv â…¢ cells including S100A11,Peroxiredoxin1, Tropomyosin3isoform2, Nucleophosmin1isoform2, S100A11andCofilin-1.S100A11and Cofilin-1can enhances migration and invasion of hepatocellularcarcinoma cell. The relationship between S100A11and EGFRvâ…¢ was significant Statistics(P=0.0282).The relationship between Cofilin-1and EGFRvâ…¢ was no significant Statistics(P=0.3662). Conclusions EGFRvâ…¢ promotes hepatocellular carcinoma cell migration andinvasion is related S100A11up-regulation. |
