Objective:To investigate the effects of cholangiocarcinoma cells (QBC939) using fluorouracil and mitomycin on proliferation, senescence, apoptosis, autophagy, and changes after apoptosis, autophagy was blocked.Methods:after respective Using fluorouracil of25-200μg/ml or0.25-2.0μg/ml mitomycin C on cholangiocarcinoma QBC939cells24h, In order to determine the combined chemotherapy drug concentration by detecting of cell proliferation with Cell Counting Kit-8, CCK-8; Detect apoptosis with Annexin V-FITC staining and flow cytometry Cellular, Observe the cellular senescence with Cellular Senescence Assay staining Kit. Single-dansyl cadaverine staining and flow cytometry to detect autophagy;3-methyl adenine or caspase inhibitor Z-the VAD-FMK to blocke the autophagy and apoptosis, and to detect proliferation,apoptosis, apoptosis and autophagy using the same methods.Result:Proliferation was significantly inhibited when fluorouracil 100μg/ml, mitomycin1.0μg/ml,And compared to no chemotherapy drugs, Inhibitory Effects of Proliferation, there was a significant difference between any group and the other group by analysis of variance(F=26.929, P<0.05). A significant increase in level of apoptosis, senescence, autophagy with statistical significance (P<0.01).On the one hand, under the apoptosis inhibitor, Reduction in the level of apoptosis, increase proliferation, senescence significantly increase, but no significant increase in autophagy. On the other hand, after joining the autophagy inhibitor, autophagy, proliferation, senescent cells decrease, but significant increase in apoptosis.Conclusion:Significantly reduce cell proliferation and induce apoptosis, senescence and autophagy, after combineing with fluorouracil, mitomycin on cholangiocarcinoma cells;Senescence, apoptosis, autophagy is different ways of Biological effects under chemotherapy, Autophagy is likely to be a mechanism for QBC939cells to avoid adverse environment.
