Toxicity Of Candidate Sealants For Rupture Of Fetal Membrane In Vitro

Objective:To provide safty-referrence when sealing rupture of fetal membrane after PPROM by evaluating the toxicity of3medical tissue adhesives tested on direct contact with fetal membranes.Methods:cyanoacrylate(CA group)、fibrin glue(FS group)、 collagen sponge(Coll group) were directly layed on little piece of fetal membrane,and there was an untreated piece as control group.After being incubated for24hours, the patches were fixed, embeded and sectioned for HE stainning with hematoxylin-eosin or stained for apoptotic cells with the use of TUNEL technology.The quantity of Bax and Bcl-2protein related to apoptosis were tested by Western blotting.Results:1.There was a significant ruptrue in the amnion layer treated with CA, while the amnion layer was complete in the3left groups.2.Compaired with FS group(16.0%±0.8%)、Coll group(13.0%±1.0%) and Con group(12.1%±0.8%),the ratio of apoptotic cells in CA group(51.0%±1.8%) was significantly higer (P<0.01).The ratio of Con group was lower than in FS group and Coll group(P<0.05). There was no statistical difference between FS group and Coll group (P>0.05).3. The expression of Bax in CA group (1.294±0.029) was significantly increased than FS group (0.421±0.023), Coll group (0.479±0.024) and Con group (0.287±0.017)(P<0.05). The level of Bcl-2in CA group, FS group, Coll group and Con group was (0.350±0.034),(0.406±0.269),(0.433±0.020) and (0.493±0.027) respectively. CA group has lower expression of Bcl-2than the left groups (P<0.05), also Bcl-2of FS group and Coll group were less than Con group(P<0.05). No statistical difference of Bcl-2existed between FS group and Coll group (P>0.05)Conclusions:1.According to the research, cyanoacrylate was harmful to the amnion layer and not suitable for fetal membrane repair.2.Considering of safty, it’s better to choose fibrin sealant or collagen to repair fetal membrane.
