Objective: To understand the mechanism of PSP onimmunomodulation and anti-tumor, we examined the gene expressions andrelated protein expressions on TLR signaling pathway in human peripheralblood mononuclear cells (PBMCs).Methods: The human PBMC was isolated and cultured in vitro andthen divided into PSP group and control group, both of which were inducedby PHA. Real time fluorescent quantitative PCR (Q-PCR) andwestern-blot were used to quantitatively analyze the expression of thegenes of TLR signaling pathway in human PBMC induced by PSP or not.Results: Compared with the control group, the gene levels of TLR4,TLR5,TLR6,IRAK4,TRAF6,TAK1,IKKα,JNK,P38,ERK,NF-κB and IRF5in the PSP group were significantly higher than these inthe control group(P<0.05). And the protein expression of TRAF6,IRF5,NF-κB and TAK1were obviously higher(P<0.05).Conclusions: PSP could play a role of immunomodulation byregulating the expressions of NF-κB and IRF5on the TLR in the PBMC, and might further activate MAPK ways to influence the growth of cells andthe secretion of cytokine by the activation of TRAF6. These could supplyevidences for further exploring the immunoregulatory mechanism of PSP....
